Objective to research the fertilizing capacity and the responsiveness of sperm to progesterone(P) and traditional Chinese medicine. Methods: After a swim-up,sperms were exposed to P 0.1μmol/L,0.5μmol/L,5μmol/L,10μmol/L and incubated in capacitating medium for 3h.Unfertilized oocytes which were kept at 37°C for up to 7 days after oocyte collection are used for this test. The sperm and oocytes are incubated together for 2h.Then the sperm which are binding to the oocytes are removed by repeated aspiration. The number of the sperm can be counted. Results: 10μmol/L,5μmol/L P caused a elevation of binding number of the sperm P〈0.05 but it had no effect in the groups of 0.1μmol/L,0.5μmol/L P P〉0.05.Conclusion: Progesterone of 10μmol/L,5μmol/L can enhance sperm-ZP binding capacity. The low P of 0.1μmol/L,0.5μmol/L caused no effect to sperm-ZP binding capacity. In light poor sperm patients, Progesterone can't improve the fertilizing capacity of sperm. After the treatment of traditional Chinese medicine, the quality and quantity of sperm and sperm-ZP binding capacity is improved significantly.
