Objective:To establish animal model of hyperhomocysteinemia,investigate the effects of hyperhomocysteinemia(HHcy)on endothelial funtion by observing the changes of NO,ET-1 in plasma of hyperhomocysteinemia rats.To confirm that hyperhomocysteinemia had injurious effect on endothelial function.Methods:Thirty male Wistar rats were randomly divided into two groups:Methoinine(Met)group(n=15) and control group(n=15).Met group were fed with a standard rat diet supplemented with 3%Methoinine for 1 month to establish animal model of hyperhomocysteinemia.Control group were fed with a standard rat diet for 1 month.All thirty Wistar rats were taken blood samples at 0d and 30d.The levels of total plasma Hcy were measured using enzymoimmunoassay.The levels of total plasma NO were measured using nitrate reductase test.The levels of total plasma ET-1 were measured using radioimmunoassay Then use t test to compare theire satistic difference.Results:At 30d,the levels of total plasma Hcy and ET-1 were higher than they at 0d,and the level of plasma NO was lower than it at 0d in Met group rats,the differences were statistically significant(P<0.05).But these changes did not appeare in control group rats.At 30d,the levels of total plasma Hcy and ET-1 were higher in Met group rats than control group rats and the level of plasma NO was lower than control group rats,the differences were statistically significant(P<0.05).The changes of the levels of total plasma Hcy,NO and ET-1 in Met group rats were different from it in control group rats,the differences were statistically significant(P<0.05).Conclusion:Endothelial function had impaired when hyperhomocysteinemia existed.Hyperhomocysteinemia had injurious effect on endothelial function.
