Objective:In order to investigate the expression of c-fos in intestinal tissue of rat with ulcerative colitis(UC)and study protective mechanism of xipayikuijiean,we developed an animal model of UC in rat;we construct the eukaryotic expression vector pcDNA3.1/CT-GFP-TOPO-c-fos,to further studies the pathogenesis of UC and target of xipayikuijiean with RNAi technique.Methods:64 Wistar rats were divided into five groups:control group(n=10),UC group(n=11),xipayikuijiean group(n=14),5-ASA group(n=14),NS group(n=15),UC rats induced by DNCB and acetic acid.Observe and assay the expression of c-fos by RT-PCR and Western blot.Construct the recombination pcDNA3.1/CT-GFP-TOPO-c-fos,after identification,transduct into COS-7 by Lipofecta-mine 2000.Observe and assay the expression of c-Fos-GFP fusion protein by inverted fluorescence microscope and Western blot.Results:Pathological section of colon showed that there were ulcerations in model group;Expression of c-Fos remarkably increasd in intestinal tissue of rats in UC group(P<0.01)as compared with control group;but it remarkably decreased after theraphy in xipayikuijiean group(P<0.05)as compared with UC group;Expression of c-Fos remarkably increasd in NS group as compared with xipayikuijiean group;expression of c-fos mRNA is no difference in five groups.Obtained encoding sequence of c-fos was identitical with that included in GenBank,and the eukaryotic expression vector pcDNA3.1/CT-GFP-TOPO-c-fos was constructed correctly.Expression of c-Fos-GFP fusion protein on COS-7 cells was proved by inverted fluorescence microscope and Western blot.Conclusion:It demonstrated tentatively that c-fos had an effect on UC and target of xipayikuijiean may be c-fos.The eukaryotic expression vector pcDNA3.1/CT-GFP-TOPO-c-fos was constructed success-fully,which will contribute to further studies on c-fos and roles of c-fos in the pathogenesis of UC.
