| Object:The study in rats was performed to observed endotoxin-induced myocardiac tissue injuries and assessed the serum concentration of cTnI and CK-MB during endotoxemia.Method:in vivo experiments,rats were randomized into seven group:NS group,LPS 0.5 hour group,1 hour group,2 hours group,3 hours group,12 hours group and LPS 24 hours group.Each group had eight rats.LPS group administrated to endotoxin(5mg/kg,dissolved by 1ml NS,ip)and NS group controlled with 1ml NS(ip).After treatment,3ml abdominal aorta blood were collected to measure serum cardiac troponin-I(cTnI)and Creatine kinase-MBmass(CK-MB);transmural blocks of myocardial tissue from the middle portion of the left ventricular free wall were collected and fixed in formalin.Then myocardiac tissue injury was observed using electron microscopy and light microscopy.Result:The myocardiac tissue injury was observed by electron microscope in LPS0.5h group;and it can be observed by light microscope in LPS1h group;with time go by,the pathologic damage evolving.The serum concentrations of cTnI and CK-MB increased from LPS 0.5h group to LPS 24h group,With time go by,the concentrations advancing;but,serum cTnI and CK-MB concentrations in LPS0.5h group have no significant higher than those in NS group(P=0.932,P=0.538),serum cTnI concentrations prominently increased in LPS 3h group(p<0.05)compared to that of NS group,serum CK-MB concentrations were significantly increased in LPS1h group compared to that of NS group(P<0.05).Conclusion:This study observed the earliest damage of endotoxin to rat myocardium appeared after 0.5 hours of endotoxin injection,but in 0.5h serum cTnI and CK-MB concentrations have no significant change.The serum concentrations of cTnI significantly increased after 3 hours of endotoxin injection, CK-MB were significant increased after 1 hours of endotoxin injection.With time go by,the damage become more and more serious.The serum concentration of cTnI and CK-MB increased in endotoxemia rat... |
