Objective:study for the interference effective of variable tidal volume(TV) on ALI rabbit model,study IL-10's controlling role on ALI rabbit model when ventilation.Methods:24 New Zealand rabbits were made into ALI model by alveolar wash(AW)ways,and then were randomized to divided into 4 groups in average to receive mechanical ventilation with either(A)V(T)=6ml/kg ideal body weight,(B)V(T)=10ml/kg ideal body weight,(C)V(T)=20 ml/kg,(D)VT=10ml/kg ideal body weight and to receive injection of 2μg/kg rhIL-10 in the same time.when ventrilated,the FiO2 was 1.0,the rate of I and E was 1:2,the frequency of breath was 34 times per minute o After ventilated for 60 minutes, A,B and C group received intravenous injection of 1ml/kg normal saline,while the D group were injected 2μg/kg rhIL-10.Bronchoalveolar lavage fluid of all animals were collected after injured(I1 point)and 60 minutes after ventrilate(I2 point),and the density of IL-10,IL-6,IL-ST,TNF-αwas detected by ELISA; blood The serum of all animals was treated by the same ways respectively before injured(T0),afterjnjured(T1),when ventrilated for 30 minutes(T2) and 60 minutes(T3),and the density of LDH was also detected by colorimetric method.Then left lower lung of all animals were taken out for pathological section and homogenate detection. Results:1.Successful ALI model was builded,and all experimental rabbits lungs showed pathological change.2.The expression of LDH in serum is decreased after ventilation on C,B,A group were in order from the top to the lower expressing level of LDH in serum.while there was no significant difference of LDH among all groups in lung tissue bomogenate.3.The expressing level of TNF-a in serum of all rabbits decreased after vetrilated;It was same to IL-8 of serum and bomogenate in A,B,Dgroup,while that in C group presented ascensus tendency.C,B,A group were in order from the top to the low in expressing level of TNF-α,IL-8,IL-6 in serum.And IL-10 of lung tissue bomogenate in all groups at I2 point expressed more than that at I1 point(P<0.01),there was no statistical difference between every two groups (P≥0.05).4.IL-10 is expression after 30 min's ventilation,and its expression is hysteresis,also the expression of IL-10 is lower than TNF-α,IL-8,IL-6 at the same point in each group,the expression of IL-10 in BALF and tissue homogenate is lower than IL-8,that is to say,IL-10 is expressed at low concentration in inflammatory.Conclusion:1.Make ALI rabbit model by lung lavag,and all the rabbits' lung tissues are injured with different degree.2.Low Tidal Volumes within safety scope,could deregulate the expression of Inflammatory cytokines,relieve the lung tissue damage greatly,high pressure or high tidal volumes interference could not only release the lung tissue damage but also aggravate lung tissue damage. 3.IL-10,as an anti-inflammatory factor,could inhibit the release of proinflammatory factors TNF-α,IL-8,IL-6,relieve Inflammatory reaction,and then reduce the degree of lung tissues' injury.
