Objective.To study the effects of siRNA(Short interference RNA) on the expression level of alpha globin chain mRNA and the proliferation ability in K562 cells at 24,48,72,96h after the transient transfection of siRNAs which were designed specifically according to the sequence of alpha globin chain with the help of Oligofectamine;to evaluate the effects of siRNA on the expression of alpha globin mRNA and proliferation in K562 cell line.Methods:The siRNAs which were chemical synthetic were designed specifically according to the sequence of alpha globin chain.Then transiently transfected the siRNAs into K562 cells with the help of Oligofectamine.Detected the expression level of alpha globin mRNA and the proliferation ability of K562 cells by Real-time quantitative reverse transcription polymerase chain reaction(RQ-PCR)and CCK-8(Cell counting kit-8)respectively at 24,48,72,96h after transfection.Results:At 48h after the transfection,the -△△Ct value of the alpha globin chain was -2.6±0.8;at 72h after the transfection,the -△△Ct value of the alpha globin chain was -1.9±0.6.The expression of alpha globin mRNA in the experimental group was reduced obviously compared with the control group(P<0.05).At 48,72,96h after the transfection,the proliferation of K562 cells in the experimental group was inhibited obviously compared with the control group(P<0.05).At 24,48,72,96h after the transfection,the proliferation of K562 cells in the Oligofectamine group was not inhibited obviously compared with the control group(P>0.05).Conclusion:The expression of alpha globin mRNA in K562 cells which were transiently transfected of the alpha globin specific siRNAs with the help of Oligofectamine was reduced effectively in 48,72h after the transfection,and the proliferation ability of K562 cells changed a lot.We proved that there was a RNA interference mechanism about the expression of the alpha globin mRNA which existed in K562 cells,the expression of the alpha globin mRNA in K562 cells could be reduced effectively by the alpha globin specific siRNAs, and the proliferation of the K562 cells was related to the expression level of the alpha globin mRNA to some extent.
