The Effect Of Exogenous Monocyte Chemoattractant Protein-1 On Bone Defect

Objective The experimental study was carried out to prove the effect of exogenous monocyte chemoattractant protein -1 on bone defect.Methods Bone defect were established in the middle of left radii of SD rats.Eighty mature male SD(Sprangue-Dawley) rats having a mean body weight of 257.8g were fed on an ordinary laboratory diet and were randomly divided into four groups: control group , group defect of bone and groups dealed with MCP-1 of different dose.5 rats from each groups were killed at 7th,14th,28th,60th days after operation. The activity of alkaline phosphatase (ALP) , the content of calcium and phosphorus in the serum, and radiography,histological were detected respectively.The 60th-days radii sample were tested biomechanically.Results The serum calcium, phosphorus and ALP level were higher in MCP-1 injected groups than those in the controls at 7th,14th,28th,60th days after operation(P<0.05). Quantitative analysis of histological examination showed that the sum of cells contained bone morphogenetic proteins-2 were greater for the experimenter groups than those of the controls at the same times(P<0.05). the sum of calluses were greater for the experimenter groups than those of the controls at 14th,28th days after operation(P<0.05). The biomechanical test results also showed the average peak deformation of the defect-healing site at 60th day were greater in the experimental group, but there were not significant difference(P>0.05).Conclusion At the early stage after bong defect,exogenous MCP-1 could be chemotactic for monocytes and induce the expression of BMP2, MCP-1 may play significant role in the healing of bone defect .
