A Study On Pioglitazone Inducing Apoptosis Of Carcinoma Of Prostate Cell PC-3

Objective:The paper presents results of effect and mechanism of Pioglitazone on inducing apoptosis of Carcinoma of Prostate Cell PC-3,which will provide a new way for therapy of Carcinoma of Prostate.Methods:Cultures of Carcinoma of Prostate Cell were exposured to Pioglitazone at the different concentrations of 0μmol·L-1,0.1μmol·L-1,1μmol·L-1,10μmol·L-1,100μmol·L-1 respectively.The effect of Pioglitazone on inhibiting hyperplasy and on inducing apoptosis were determinated with MTT,Tunnel.The activity of Caspase-3 were determinated with flow cytometry.Rusults:The hyperplasy of PC-3 were significantly inhibited in the 10μmol·L-1,100μmol·L-1 groups, but the effcect was nonsignificant difference in 0.1μmol·L-1,1μmol·L-1 groups. PC-3 were significantly induced by Pioglitazone in the 10μmol·L-1,100μmol·L-1 groups.The effects in 0.1μmol·L-1,1μmol·L-1 group were nonsignificant difference.The activity of Caspase-3 in 10μmol·L-1,100μmol·L-1group were significantly higher than other groups, The activity of Caspase-3 in 0.1μmol·L-1,1μmol·L-1groups were a litter high than control group.But The expression of active Caspase-3 was nonsignificant difference in 0.1μmol·L-1,1μmol·L-1 groups.Conclusions:The Apoptosis were induced by Pioglitazone on Carcinoma of Prostate Cell PC-3 in a dose-dependent manner.The mechanism were that the high activity of Caspase-3 were induced by Pioglitazone,which induced apoptosis of PC-3.
