Objective To investigate the effects of 1, 25—dihydroxyvitamin D3 on limballograft and the feasibility of 1, 25—dihydroxyvitamin D3 combining FK506 inpreventing allograft acute rejection in rat limb transplantation.Method After theestablishment of rat limb allograft, inbred strain male Wistar ratlimb were transplanted to male SD rats. The rats were divided into 4 groups randomly. Afteroperation SD rats were treated with physiologic saline in groupe A, The SD rats wereconditioned with. 1,25—dihydroxyvitamin D32.5μg·kg-1·d-1, FK506 po 1mg/kg every day separately or unitedly in groupe B, CandD. After operation, Generalstate were observed in all groups. Peripheral T lymphocytic subsets were determinedon d1,3,7,14,28posttransplantation and pretransplantation. The histologic grading ofskin rejection of limb allograft and the hepatic and renal function were determinedposttransplantation.Results The mean survival time (MST)of limb allografts was prolonged from(12.250±0.375)days to (8.500±0.500) days by treatment of the recipient rats with1,25—dihydroxyvitamin D3. The MST of limb allografts in group VD3+ FKS06 waslonger than in group FK506. Peripheral T lymphocytic subsets and The histologicgrading of skin rejection of limb allograft were the same condition as the MST oflimb allografts in the corresponding group P<0.05.Conclusions 1,25—dihydroxyvitamin D3 can relieve allograft acute rejection and thedamage of recipients and allografts and can prolong the survival time in rat limbtransplantation. 1, 25—dihydroxyvitamin D3 in combine with FK506 has a synergisticeffect and can acheve the improvement of graft survival.
