Basic fibroblast growth factor (bFGF) is a kind of wide-spectrum mitogenic factor, bFGFhave biological effective to most mesoderm-and neuroectoderm-derived cells. It has the goodeffective to nerve preservation, myocardial preservation, avoiding anaemic as well aspromoting the rave surface heal. So its clinical treatment effective is paid close attention dayby day. In this report canine corneal epithelial cell were cultured primarily in DMEM/F-12culture medium to investigate the exogenous bFGF promoting effective. The third generationof canine corneal epithelial cells were exposed to different concentrations bFGF(lng/mL,10ng/mL, 100ng/mL, normal control group) medium, and the proliferation characteristics ofthe cells were measured with a methylthio Tetrazole(MTT) assay. Measure optical density(OD)and calculate relative growth rate after 24h, 48h and 96h. Result: OD in each group wasincreased with the culture time longer and the concentration of the bFGF higher. The OD ofcomeal epithelial cell cultured on the medium contain bFGF were significantly higher than inthe control group(P<0.05). OD increased from 1ng/mL, and reached the peak at 10ng/mL andthen decreased gradually. Conclusion: bFGF can promote proliferation of canine comealepithelial cell when concentration higher than 1ng/mL. The capability was correlated withconcentration in certain field. 10ng/mL was the best dose because OD in this point werehigher than other groups at 24h, 48h and 96h.Besides, we set up the 6mm diameter defect model using trephine on canine corneal toanalyses the treatment effective of bFGF on canine vivo. The twenty four mongrel dogs weredivided into four groups randomly: PBS group, 0.1μg/mL bFGF group, 0.5μg/mL bFGFgroup, 1μg/mL bFGF group. The canines were applied with designed eye drops, 3 times daily,two drops every time. They were observed the healing procedure through received fluorescentsolution at the same time at day 1, 3, 5 and 7 postoperatively. Pathological sections were madeto observe healing of corneal epithelial and the arrangement of collagen fiber. The resultindicated that the disparity was extremely signification on the first day after surgery while theheal ratio of 0.5μg/mL bFGF group was 27.47%, 1μg/mL bFGF group was 31.23% contrastthe control group(P<0.01). At the third day and the fifth day, the ratio of bFGF group werehigher than control group(P<0.01).Most case were recovery except two cases in control group on seventh day. The study demonstrated that the addition of bFGF to corneal woundsresulted in a significant increase in the extent of re-epithelialization and the recovery offibroblast. Did not find bFGF can effectively promote or restrain the growth of the cornealneovascularization.
