[OBJECTIVE] A study for Chitosan(Chi) to influence the expression of apoptosic gene in human umbilical vein endothelial cells in ordre to find a new stent coating substance .[METHODS] We put Chitosan with different concertration into substrate to culture the human umbilical vein endothelial cell lines.Experimental groups were classified as following:Chi group:①ethanoic acid control group(Chi 0mg/ml)②Chi:0.01mg/ml③Chi:0.1mg/mg④Chi:1mg/ml;.After incubating 48 hours,we detect a few indexes with different assay:cell activity by methyl thiazolyl tetrazolium (MTT),endothelial cell apoptosic rate by FCM,the Bcl-2 and Bax protein expression by Immunocytochemcal (IHC),cell apoptosic modality by AO,the expression of Bcl-2 mRNA and Bax mRNA by reverse transcription polymerase chain reaction (RT-PCR).[RESULTS]⑴After incubating 48 hours,endothelial cell activity increased by MTT as Chi concertration increased. The absorbency of different groups reduced as Rapa concertration increased,and consequently the endothelial cell activity decreased.⑵The HUVECs apoptosic rate decreased as Chi concentration graduately increased,the apoptosic rate of experimental groups respectively were :23.53+3.28%,20.01+2.7%,9.7+3.1%,Compared with control group(38.1±3.3%);the apoptosic rate of Chi groups have a significant difference while the apoptosic rate of Rapa groups show no obvious difference.by FCM⑶Chi could significantly increased Bcl-2 protein expression and decreased Bax protein expression as its concentration(Chi :0.01mg/ml~1 mg/ml) increased by Immunocytochemcal(IHC).⑷As Chi concentration increased could significantly increased Bcl-2 mRNA expression and decreased Bax protein and Bax mRNA expression by reverse transcription polymerase chain reaction . [CONCLUSION] Chi could obviously increased the activity of endothelial cells and decreased apoptosic rate as its concentration,Chi could significantly increased Bcl-2 mRNA and Bcl-2 protein expression and decreased Bax protein and Bax mRNA expression .
