Objective: To study if ischemic pretreatment (IP) could induce themultiplication of endogenous neural stem cells and if the treatment of HuaTuo Zai Zao (HTZZ) after IP could enhance the multiplication and relieve theimpairment of repeated ischemia.Methods: Cerebral infarct model was established in rat by twicethread-ligation method. 40 healthy male rats (body weight 250-300 gram)were divided into 4 groups randomly, sham (SS) group, middle cerebralartery occlusion (SS+MCAO) group, ischemic pretreatment (IP+MCAO)group and HTZZ (IP+MCAO+HTZZ) group, with each group 10 rats. Thedefection of neural functions of rats and pathologic change in each groupwere assessed. The quantity of neural stem cells, the expression of heat shockprotein 70(HSP70) and matrix metalloproteinase-9(MMP-9) was detected byimmunohistochemical method (SP).Result: (1)In the third day after reperfusion, the neurological scores of rats insham group, middle cerebral artery occlusion group, ischemic pretreatmentgroup and HTZZ group were (0.00±0.00), (2.70±0.67), (1.70±0.67),(1.40±0.52); In the seventh day after reperfusion, the neurological scoresof rats in sham group, middle cerebral artery occlusion group, ischemicpretreatment group and HTZZ group were (0.00±0.00), (2.60±0.52),(1.50±0.53), (1.20±0.42). There was statistical significance between eachother group (P<0.05) except the relation between ischemic pretreatmentgroup and HTZZ group (P>0.05).(2)The quantity of neural stem cells insham group, middle cerebral artery occlusion group, ischemic pretreatmentgroup and HTZZ group were (50.00±5.98), (342.10±50.99), (519.20±37.02),(591.40±31.93),with statistical significance (P<0.05 ).(3)The expressions ofheat shock protein 70(HSP70) in sham group, middle cerebral arteryocclusion group, ischemic pretreatment group and HTZZ group were(0.00±0.00), (55.70±4.79), (73.50±4.60), (92.00±4.45)(P<0.05); theexpression of matrix metalloproteinase-9(MMP-9) were (0.00±0.00),(44.70±3.86), (26.60±2.12), (18.10±2.03)(P<0.05).Conelusion: Local cerebral ischemia could activate the regeneration ofendogenous neural stem cells in cornu. Ischemic pretreatment could promotethe regeneration of neural stem cells,raise the expression of protection factor,decrease the expression of impairment factor and relieve the injury ofcerebral ischemia. HTZZ could enhance all the more.
