Study Of Quantitative Analysis Of Aristolochic Acid In Chinese Herbal Medicine And Its Preparations

Chinese herbal medicine has several thousand years of history , thereby creating a splendid culture of Chinese medicine. Chinese herbal medicines are not universally recognized by the world, the main reason is that Chinese medicine ingredients is too complex, it is not to explain about their precise composition and effective pharmacological effects, there are not reliable quality standards to follow sometimes.In recent years, the aristolochic acid induced ulcers and aristolochic acid nephropathy , the condition attracted widespread attention at home and abroad, some herbs abroad were banned.Most of determination of aristolochic acid content methods were reported in the reference., but these methods is no systemic. In order to better control the quality of aristolochic acid and its preparation, in this paper a broader applicability of aristolochic acid content determination method was establishded in accordance with reference. Current determination of aristolochic acid content methods are TLC, HPLC, LC-MS and so on. HPLC was the most widely used,the most reference substance are aristolochic acids I (A)and II (B). On the subject, TLCS and HPLC lowest detectable limit was inspected . The results showed that HPLC detection limit is less than TLC method detection limit. So in this subject , determination of aristolochic acid content method by HPLC is focal point.Based on the reference, we developed a HPLC method to determine the content of aristolochic acid. The HPLC system using a mobile phase of 0.3% ammonium carbonate solution-acetonitrile (75:25, v/v) with pH 7.5 in combination with a C18 column was useful for determining aristolochic acids I.The detection wavelength was set at 250nm, extraction solvent is methanol-formic acid (98:2), extraction methods is heating regurgitation two hours.In methodological study, followed 2005 version " Pharmacopoeia of China " of a new appendix to the "Chinese medicine quality standard analytical method validation guiding principle", we inspected the aristolochic acid A linear range , it was good linearity when the mass is within 1.0224 ng and 511.2 ng,the correlation coefficient was 1.0000, and the repeatability was 100.9%.The method is rapid,accurate,simple,suitable and durable.The collection of the 22 samples of Chinese herbal medicine Aristolochia and Asarum were identified, and then the establishment of HPLC method was to determine them . Determination of the herb Asarum caudligelellum C.Y.cheng et C.S.Yang and Asarum sugittarioides C.F.Liang,2 kinds of medicines have statutory standards Aristolochiaceae.Determination of the medicines were not reported in the reference.The research finding was to provide the scientific basis in Aristolochiaceae Chinese herbal medicine quality standards.it is an important value.
