BackgroundGlomerular mesangium is composed of glomerular mesangial cell (GMC) andextracellular matrix (ECM). The pathologic change of glomerular mesangialis the basiswhich leads to glomerular sclerosis and renal failure. GMC proliferation (during variesof immune and non-immune injuries) lead to aggradation of big molecular substance onglomerular mesangium and the mechanical obstruction on the glomerular capillarieswhich reduce the glomerular filtration rate (GFR).How to control the GMCproliferation has great significance in reversion of glomerular injury and chronicchange of glomerular disease. ACEI protective effect of kidney is hotspot of research.plays an important role in protecting the kidney. The experiment reveals the ACEIaffection on the GMC proliferation and ACEI protective effect of kidney and itsmechanism. A great many of researches reflect varies of factors (both the promotingand inhibiting factors) affect the GMC proliferation. TNF-αpromotes the GMCproliferation and IL-10 inhibits it. So we study the expression of IL-10 and TNF-αin GMC and discuss the ACEI effect on GMC proliferation.ObjectiveObserving the conveying of IL-10 and TNF-αon GMC under the differentconcentration of ACEI.So to study Whether ACEI can inhibite the GMC proliferationor not.MethodCultrued rat GMC were incubated with different concentration FOS and sameconcentration LPS for 48 hours.Then they were dyed with the SABC immunohis tochemistry reagent, IL-10 and TNF-αAb.Computer image-analyzing system was usedto analyze those GMC slices.Statistical TreatmentAll data have been analysed statistically. Measurement data was expressed by means of (?)±s.SPSS11.5 software was used for comparison, f-test to test interclassdifference, P<0.05 was significant.Results1. The dyed-positive cells of IL-10 and TNF-αshow some brown particles.2. For IL-10group, the more FOS, the more particles.3. For TNF-αgroup, the more FOS, the less particles.Conclusion1. There is inhibition of FOS on the GMC proliferation.2. There is dose-dependent trend of IL-10 and TNF-αinhibition on GMC proliferation.
