Objective and Significance: The hepatocyte growth factor(HGF) is a multifunctional cytokine and its biologic activity is mediated by C-Met.The growth,invasion and metabasis of many human tumer cells are closely in relation to HGF/C-Met. The invasion and metabasis are the main cause of the laryngeal carcinoma patients'death. We detected and analysed the expression of HGF and C-Met in the polyp of vocal cord ,the larynx squamous cell cancer (LSCC), the lymph node of neck, and the expression of HGF in serum in order to study 1).The variabilit of the expression of HGF and C-Met in the polyp of vocal cord and the LSCC.2). The variabilit of the expression of HGF and C-Met in the Lymph node of neck before and after metastasis.3). The relationship between the expression of HGF in serum and the malignant extent,clinical stage,metasis,prognosis.Thus,we study the function of HGF/C-Met in the genesis and advancement of the laryngeal carcinoma as well as the relationship between HGF/C-Met and the prognosis of the laryngeal carcinoma.Methods: 36 patients with laryngeal carcinoma includes:12 patients with the metabasis of lymph node of neck,;25 polyp of vocal card patients(rule out malignant tumor and other major diseases) were choosed as controls. The expression of HGF and C-Met in the polyp of vocal cord ,the larynx squamous cell cancer (LSCC) and the lymph node of neck were detected by the immunocytochemistry ,in addition to the expression of HGF in serum was detected by ELISA.The patients'clinical materials considering pathological class, clinical stage ,metabasis were collected.Also follow up the recurrence and the metabasis of the laryngeal carcinoma patients after operation. All the data were analysed by SPSS statistic tool.Results:1. In normal membrana muCosa laryngis ,there were poor protein expression of HGF and C-Met.2. In CanCer of larynx,expression of HGF and C-Met was signifiCantly Correlated with surgiCal stageⅢandⅣ,histologiC grade 3 ( P < 0. 05) . But was not Correlated withage ,invasion and lymph metastasis( P > 0. 05) . HGF expression was signifiCantly Correlated with surgiCal stageⅢandⅣby semequantitative and analysis.3. The content of plasma HGF in patients with cancer of larynx was nearly four times higher than that of the healthy men , and the content of serum HGF decreased significantly after surgery. Correlation analyses showed thatthe HGF level was correlated with tumor size ,vessel involvement ,node status ,and TNM stage. No significant associations were found between the serum HGF content and age ,menopause status ,estrogen eceptor , progesterone receptor .4. After operation ,the content of plasma HGF in patients with cancer of larynx markedly decreased(P<0.05)5. The content and alleosis of plasma HGF in patients with cancer of larynx correlates with the recurrence and the metabasis of the laryngeal carcinoma patients after operation.Conclusions:1. The expression of HGF and C-Met correlates with the advancement of the laryngeal carcinoma。2. The high expression of HGF and C-Met correlates with the genesis of the laryngeal carcinoma.3. The concentration of circulating hepatocyte growth factor may act as an independent prognostic indicator in primary cancer of larynx.
