PART ONE Experimental Study On Superparamagnetic Iron Oxide (Feridex)Labeled Breast Cancer Stem CellsObjetive: To study superparamgnetic iron oxides(SPIOs) labeled breast cancer stem cell s (BCSCs) .Methods: Feridex (one kind of SPIOs) and Poly-L-lysine were added into the medium to be co-cultured to make magnetic labeled BCSCs. With transmission electron microscopy (TEM), Prussian blue staining and MRI in vitro ,we identify the Feridex-labeled BCSCs.Results: When BCSCs incubated with Feridex-PLL, we can see the iron particles are in intracellular (non-nucleus) and also existed in dauhgterclones. Feridex-labeled BCSCs are in low signal in MRI. And with concentration past, Magnetic labeled BCSCs show different signal intension .Conclusion: We successfully labeled BCSCs with Feridex to be magnetic labeled BCSCs. And we were also observing magnetic labeled BCSCs in vitro with MRI. With concentration past, Magnetic labeled BCSCs show different signal intension .PART TWO Experimental Study on the biology characteristic of superparamgnetic iron oxides(Feridex) Labeled Breast Cancer Stem CellsObjective: To study the biology characteristic of superparamgnetic iron oxides(SPIOs) labeled breast cancer stem cells (BCSCs) . Methods: Feridex-PLL complex labeled BCSCs were use to be magnetic labeled BCSCs. According to concentration of Feridex cells divided into three groups: 15μg/ ml group,25μg/ ml group,35μg/ ml group, control group is unlabled cells. The biology characteristics of labeled BCSCs were examined with MTT.Results : When BCSCs were incubated with Feridex, With the concentration of Feridex increased 15μg/ ml-25μg/ ml ,compared with control group here was no obvious difference of the growth and proliferation of BCSCs ( P > 0. 05) . When the concentration of Feridex increased 35μg/ ml , there existed an obvious difference ( P < 0. 05) . Conclusion : BCSCs are successfully labeled with Feridex that is a kind of SPIOs to be magnetic labeled BCSCs. That is the base of tracking magnetic labeled BCSCs in vivo with MRI.
