Objective Fibroblast growth factors and their receptors(FGFs/FGFRs) exist in skull,organization(lung,kidney,skin),skeleton etc,and they play an importent role in skeleton growth and the union of fracture.Many development diseases and some delay-union or disunion of fracture have a compact relationship with FGFs/FGFRs. We take advantage of transgenic mice to make fractured model where the low expressed FGFR2 can only be existed in the osteoblast .we forge on this new method to explore the effect and mechanism of FGFR2 in the union of fracture .so we can make problem in the union of fracture more clearly and supply preferable rationale for the clinicMethod We make the OC-Cre mice who express Cre recombinant protein only in osteoblast and FGFR2-RNAi mice cross,and then get their offspring FGFR2RNAi;OC-Cre mice.The new mice have a special feature :only have a low expression of FGFR2 in osteoblast.we make the fracture model and observe the union of fracture.1 global morphous 2 X-ray 3 histological section . Result1. global morphous of FGFR2RNAi;OC-Cre mice are smaller than controls,P<0.001;2.X-ray The fracture of wild tipe(WT) are to heal 21days after operation but the FGFR2RNAi;OC-Cre are not until 28 days.So the FGFR2RNAi;OC-Cre mice are transparently slow than the WT.3.histological section FGFR2RNAi; OC-Cre mice get few bony callus than the controls;we take advantage of a specific stain to compute the cartilage areas in different time and find that the pick value of FGFR2RNAi;OC-Cre mice are slow and small than the controls,P<0.001.Conclusion That the FGFR2RNAi;OC-Cre mice get a quite low speed in the union of tibia fracture than controls supply FGFR2 play an important role in the union of fracture.If it is lost or have a low expression ,the bone fracture may get a late healed or disunion.
