OBJECTIVE To test the expression of EphA2 and nm23-H1 in chronic cervicitis, cervical intraepithelial neoplasia(CIN) and cancer of the cervix, exploring the relationship between the expression of EphA2 and nm23-H1 and biological behavior and prognostic,and offer theoretical evidence to prognostic decision.METHODS SP immunohistochemistry stain of EphA2 and nm23-H1 was carried out to for paraffin specimen from 100 patients with cervical cancer including 18 with adencarcinoma and with squamous carcinoma, other 25 patients with cervical intra-epithelial neoplasia(CIN) , 10 with chronic cervicitis. The relationship between the both factors with the patient's age, cervical pathologic diagnosis, pathologic classified, FIGO stage, and lymphonode metastasis was analyzed. 50 patients with cervical cancer were followed up for up to two years to research the relation between EphA2 and nm23-H1 with prognosis of these patients.RESULTS 1 EphA2 and nm23-H1 both expressed in chronic cervicitis, cervical intraepithelial neoplasia(CIN) and cancer of the cervix, EphA2 protein expressed in cellular membrane and plasma, nm23-H1 protein expressed only in cellular plasma2 Positive rate of EphA2 was significantly higher in cervical cancer than that in CIN and chronic cervicitis, which was 88%,48%,30% respectively(P<0.05),and the positive rate had no relation with patients age, FIGO stage or histologic classification, but significantly related to pathologic class and lymphonode metastasis. 3 Positive rate of nm23-H1 was 100% and 84% in chronic cervicitis and CIN respectively, much higher than that of 39% in cervical cancer(P<0.05), in which adencarcinoma was higher than squamous carcinoma with no statistic value. the expression of nm23-H1 in cervical cancer was related to FIGO stage and lymphonode metastasis. 4 EphA2 had negative correlation with nm23-H1 in cervical cancer(r=-0.51). 5 Follow-up for 50 patients with cervical cancer showed that patients with positive EphA2 and negative nm23-H1 had worse prognosis than other groups.CONCLUSION Expression of EphA2 and nm23-H1 have relationship with cancer invasion , metastasis and prognosis for patients with cervical cancer.and these two factors can act as a prognostic index for patients with cervical cancer.
