Objective: To establish the rat model of heart deficiency pattern ,and to discuss the relationship between heart defiency pattern and liver, lung and brain.Methods: The rat model of heart deficiency pattern was established by using flower pot technique with alarm clock depriving rat REM sleep for 96 hours and the model was evaluated. Morphology changes of heart, liver and lung were observed by regular HE s. technique; Activity changes of glycogen, AKP and SDH in lung were observed by histochemistry methods; Content changes of SDH in liver mitochondria and COX in brain mitochondria were detected with colorimetric method andoxidation electrode method.Results:96 hours after the establishment of the models, heart rate of rat, respiratory frequency, systolic pressure and exhausted swimming time showed significant difference comparing with the control group (P<0.05); The ultrastructures of cardiocytes were damaged and the mitochondria reduced; the regular HE s. showed that the morphology changes of the rats'organs in the early stage of modeling were not obvious;The histochemistry staining showed that the positive reactant changed obviously;The activities of 3 detection indexes were evidently lower than those of the control group (P<0.05);The SDH in liver mitochondria and the COX in brain mitochondria of model group decreased obviously compared to those of the control group(P<0.05).Conclusion: Conclusion: The models established by the method of this experiment accorded with the criteria of heart deficiency pattern. The liver, lung and brain tissues of heart-deficiency-pattern models were damaged to a certain extent. |
