Objective: To investigate the expression of Numb in fibrosis peritoneal tissue by establishing peritoneal fibrosis model. Methods: Sixty male SD rats weighing 180-220g were randomly divided into three groups which were normal control group, fibrosis two weeks model group and fibrosis four weeks model group. The peritoneal fibrosis rat model was established by intraperitoneal injection of 4.25% peritoneal dialysate fluid per day. Twenty rats of every group were sacrificed respectively at 14d and 28d. The changes in respect of histomorphology and pathology and peritoneal equilibration of function were detected in order to prove successful peritoneal fibrosis model. Expression of Numb mRNA and protein was detected by RT-PCR, Western blot in fibrosis peritoneum. Results: Expression of Numb mRNA and protein in the fibrosis model group was decreased (P<0.05), and expression of E-cadherin in the fibrosis model group was also decreased (P>0.05).However, expression of Col-I andα-SMA in the fibrosis model group was increased (P<0.05). Conclusion: Numb induces directly or indirectly Epithelial Mesenchymal Transition in peritoneal mesothelial cell.
