Objective To explore the effects of different dose of Propofol on spatial learning and memory and hippocampus of the rats.Methods:forty eight male Wistar rats(weighted 50±7g),were randomly divided into four groups(n=12,each)as follow:A 0.9%normalsodium 10ml/kg; B,Propofol 50mg/kg;C,Propofol 100mg/kg;D,Propofol 200mg/kg.Propofol were injected continually in five days.Every day Morris water maze were used to detect the study and memory ability.After the test,each group randomly select six rats to detect the p-CREB expression of hippocampus of rats by iminunohistochemistry;the rest rats were used to observed the ultrastrcucture features of hippocampal synapses and neuron cell by transmission electron microscope.Results:compared with A group,the number of p-CREB protein expression in hippocampus in C,D group was significantly down-regulated.The result of transmission electron microscope showed that:the vacuolization of nerve cellularedema was appeared,and vacuode also appeared in the bioblast.Some of the mitrochondial crista was degranulated,and the structure of the synaptic injured.Conclusion:High dose of Propofol injure nerve cell,effect synaptic structure, decrease the express of p-CREB,so it is effect learning and memory.
