Expression Of PRL-3 Phosphatase In Human Gastric Carcinomas And Effects Of Its Specific SiRNA On Cellular Proliferation And Motility

Objective To study the expression of liver regeneration of phosphatase -3 in gastric carcinoma and the effect of small interference RNA on PRL-3 mRNA of gastric cancer cell SGC7901.Methods 59 gastric cancer tissues and 14 tissues near gastric cancer was collected in oncology department of the union hospital affiliated of Fujian Medical Unveisity in 2004,and the expression in protein level of PRL-3 was tested by immunohistochemistry,and the relation between PRL-3 and clinical pathology characteristic of gastric cancer was analyzed;Specific PRL-3 siRNA was synthesized by transcription in vitro;The role PRL-3 plays in the growth of gastric cancer cells which were transfected by siRNA through oligofectamine was detected by Methyl thiazolyl tetrazolium (MTT) assay and the most suitable transfection concentration was screened out; The expression of PRL-3m RNA in SGC7901 which was transfected 48 hours was tested by RT-PCR; Motility and migrating velocity of cells was tested by wound healing assay.Results The postive rate of PRL-3 was significantly higher in gastric cancer than in normal tissues (P=0.020 ), and expression level of PRL-3 was related to infiltrating extent(P=0.020), lymphnode metastasis(P =0.002) and TNM stage (P =0.002), but has not relation to sex( P =0.149),age(P =0.169) and histological differentiation(P =0.400);Different concentrations of siRNA can inhibit growth and proliferation of gastric cancer cell SGC-7901,especially when concentraion is 5nM;Expression level of PRL-3 mRNA was significantly lower in the group transfected into PRL-3siRNA compared to the group transfected scrambled siRNA and the blank one,but the expression ofβ-actin mRNA has no difference in the three groups;Width of wound was significant wider in the cell group transfected into PRL-3siRNA compared to the group transfected scrambled siRNA and the blank one.Conclusion PLR-3 has close relationship with gastric cancer pathology and plays an important role in incidence and metastasis process of gastric cancer;PLR-3 specific siRNA can inhibit the expression of PRL-3 in SGC7901 and proliferation and migrating of cells.
