Objective To investigate the effects of captopril on the expression of carnitine patmitoyl transferase -1 and glucose transporter-4 during the development of atherosclerosis(AS) in rabbits。Methods Three groups of rabbits were studied: group A (n=8), control rabbits; group B (n=9), HC-diet rabbits(include of 5% peanut oil,5% pork fat,15% egg yolk,0.5% cholesterol; group C (n=9), HC-diet rabbits accompany with captopril (25mg/kg.d). we detected serum TC,TG,LDL with automatic biochemistry analyzer;Histological changes in ascending aorta were observed by HE-staining and histomorphometric analysis. The gene expression of carnitine patmitoyl transferase -1 and glucose transporter-4 in vascular smooth muscle layer of thoracic aorta was evaluated by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR)technique.Resultsâ‘ At the end of 9th,25th week, TC,TG,LDL in group B and C were higher than those of group A(P<0.05),and in group B and C,TC,TG,LDL at the 25th week were higher than those of 9th week(P<0.05).â‘¡Pathomorphology indicated that extensive aortic intima(I),intima and media(I + M)became thickened,and the ratio of the area of intima to media(SI / SM)was increased.â‘¢Aortic intimal proliferation in HC-diet group was associated with a marked decrease in CPT-â… mRNA expression and increase in GLUT-4 mRNA expression in vascular smooth muscle layer of thoracic aorta . captopril attenuated artery intimal proliferation and markedly increased the enhanced CPT-â… mRNA expression and decreased in GLUT-4 mRNA expression level in vascular smooth muscle layer of HC-diet rabbits.There were significant differences of CPT-â… expression and GLUT-4 expression among the three groups(P<0.05).Conclusions Early treatment with captopril had a crucial inhibited genesis and development of atherosclerosis,induced by up-regulating CPT-â… expression and down-regulating GLUT-4 expression in vascular smooth muscle layer.
