Expression of INH receptors and ACT receptors genes in normal human ovaryObjective: To explore the relationship between INH receptors,ACT receptors genes expression in normal human ovary and serum E2,T,FSH,LH,INHB levels.Methods: Real time PCR was used to determine the ratios of the expression of INH receptors genes,ACT receptors genes to GAPDH gene. Serum E2,T,FSH,LH,INHB levels were determined by a solid quantitative sandwich enzyme immunoassay technique(Sandwich ELISA), the correlations between each gene and each hormone were analyzed.Result:①in the group of the follicular phase, INH receptorβ-glycan,ACT receptors ActRIA,ActRIB,ActRIIA,ActRIIB genes expression were higher than those in the phase of the luteinic period;②INH binding protein/P120 gene expression was very low in normal human ovary, with almost same levels in two groups;③in the group of the follicular phase, INH receptorβ-glycan gene expression was positively correlated with serum E2,FSH,LH,INHB levels.Conclusion: INH receptorβ-glycan and ACT receptors genes expression were relatively high in follicular phase of normal ovary. INH binding protein/P120 gene expression was extremely low in normal ovary. INH binds INH receptorβ-glycan ,increases its binding ability to ACT type II receptor , can mediate functional antagonism of ACT. INH - ACT - FS system especially inhibin and inhibin receptorβ-glycan were closely related with follicular development. The levels and significance of INH receptors and ACT receptors genes expression in human ovaries of patients with polycystic ovary syndromeObjective: To evaluate the genes expression of INH receptors and ACT receptors in human ovaries of patients with polycystic ovary syndrome(PCOS). Methods: The ratios of the expression of INH receptors genes,ACT receptors genes to GAPDH gene were determined by real time PCR. Serum E2,T,FSH,LH,INHB levels were determined by a solid quantitative sandwich enzyme immunoassay technique(Sandwich ELISA). The relationship between gene expressions and serum hormone levels in each group were analysed.Results:①M ean(±SEM) level of the ratio of INH receptorβ-glycan to GAPDH gene expression was higher (P < 0.01) in patients with PCOS(0.388±0.101) than in controls(0.259±0.104);②In PCOS women, expression of INH receptorβ-glycan was positively correlated with serum E2,FSH,LH,LH/FSH,INHB levels.Conclusion: It was suggested that the gene expression of INH receptorβ-glycan was greatly raised in ovary tissues of patients with PCOS. Inhibin and inhibin receptor action were maybe the local mechanism of the INH-ACT-FS system playing a role in PCOS.
