With the rapidening modern life step , sleeplessness increasingly seriously affects people's normal work and life . Good sleep is important for maintaining healthy body . At present , the research areas are primarily focused on cell electrophyscology, day-nigt rhythm balancing and sleep physcology and so on ,and having many theories . The paper is based on 5-HT theory , primarily studied on improving sleep quality by the compound prescription of traditional Chinese medicine.The compound prescription of traditional Chinese medicine is included: Ziziphus jujuba Mill.Var. Spinosa(Bunge)Hu ex H. F.Chou,Polygala tenuifblia Willd.,Poria cocos(Schw.) Wolf,Acorus tatarinowii Schott.,Ganoderma lucidum(Leyss.ex Fr.) Karst.,Platycladus orientalis(L.) Franco,Codonopsis tangsheng Oliv. Extractants of the compound prescription are made from podwer, and feeded on mice as a series of concentration gradients. Then observe sleep quality of mice . Sleep quality of mice is judged by the hypnotic latent period induced by sodium pentobarbital, the hypnotic lasting time induced by sodium pentobarbital, rate of sleeping mice induced by sodium pentobarbital at the critical dose . Addtionally, the volume of 5-HT and tryptophan hydroxylase (TPH) in the brain of mice judge the quality of sleep of mice .The experiment results indicate that the compound prescription may shorten longth of the hypnotic latent period induced by sodium pentobarbital, compared to control group low dose group ,middle dose group and high dose group respectively shortening 5.0% ,20.2% and 23.1%. the prescription drug can extend the hypnotic lasting time induced by sodium pentobarbital , compared to control group low dose group ,middle dose group and high dose group respectively extending 57.3%, 85.1%and 98.5%. On rate of sleeping mice induced by sodium pentobarbital at the critical dose, compared to control group low dose group ,middle dose group and high dose group respectively increasing 4.3%,14.3% and 31.0%。Chemistry spectrofluorometry assays the volume of 5-hydroxytryptamine and tryptophan hydroxylase (TPH) in the brain of mice .Then results indicate that compared to control group, the volumes of 5-hydroxytryptamine of low dose group ,middle dose group and high dose group all increase ,but the volumes of tryptophan hydroxylase (TPH) of low dose group ,middle dose group and high dose group in the brain of mice don't increase.
