Objective: To look for a simple,reliable method to establish and evaluate the model of the detrusor instability(DI) in guinea pig.In our research,we examine the morphology and distribution of interstitial cells of Cajal(ICCs)-like cells of the DI, and deduce these changes of ICCs-like cells, so as to demonstrate one of the mechanisms of DI.Methods: Ligated the perineal urethra of the adult female guinea pigs. Punctio vesicae was operated 6 weeks later, followed by a filling cystometry. And then observe the bladder pressure, capacity changing and the probability of DI.According to the results,the model of guinea pigs were allocated to DI group and detrusor stabilization(DS) group.Another some healthy female guinea pigs served as control group. The morphology and distribution of ICCs-like cells in the detrusor, which were taken from these guinea pigs, were quantitatively analyzed with frozen section and the monoclonal antibody to receptor tyrosine kinase(KIT) by an indirect immunofluorescence methods.Results:â‘ DI ratio of the experiment group was 35.9%,the control group was no DI. Compared with those in the control group,the maximum capacity and pressure of the bladder increased significantly in the detrusor instability group(p<0.01).â‘¡ICCs-like cells were identified to exist in detrusor in adult guiea pig bladder. In the detrusor, these cells were seen at the edge of smooth muscle bundles and within the bundles of smooth muscle cells,running in parallel with the bundles.These cells were spindle-shaped ,which characterized by bipolar cells.Compared with control group, DI group showed considerably increased in number of ICCs-like cells in the detrusor.Conclusion:â‘ Ligating the perineal urethra,and filling cystometry after punctio vesicae are a simple, reliable method to establish and evaluate the model of the bladder outlet obstruction in guinea pig leading to the DI.â‘¡The monoclonal antibody to KIT by an indirect immunofluorescence is the primary culture method of ICCs-like cells in vitro.â‘¢The increase of ICCs-like cells in the detrusor, which maybe lead to an enhanced spontaneous contraction of detrusor, maybe ICCs-like cells is the theoretically existing pacemaker cells,maybe one of the important mechanisms of DI.
