This paper introduced the mechanism and the therapy of benign prostatic hyperplasia(BPH), and the advance in pharmic research of Semen Sinapis Albae. It was a study on theextraction, separation, and content determination of the main chemical components inSemen Sinapis Albae. It discussed the inhibition of the extractions from Semen SinapisAlbae on BPH. It also studied the effect of extractions from Semen Sinapis Albae on thegrowth factors and androgen receptor. The experimental methods and results were asfollows.Sinalbin, sinapine, [3-sitosterol and volatile oil were seperated from Semen SinapisAlbae. The content of sinalbin was determinated by titration of BaSO4 precipitation. Thecontents of sinapine andβ-sitosterol were determinated by HPLC. The chemicalcomponents and the relative contents of the chemical components of volatile oil wereanalyzed and determinated by GC-MS.Using animal experiment of mice prostatic hyperplasia induced by exterior hormone,to study that the extractions could inhibit mice prostatic hyperplasia, then taking thegrowth factors and AR as research object,to study the molecular mechanism of the extractionsinhibiting mice prostatic hyperplasia. The results indicated that: (1) The extractions couldsignificantly reduce the weight of prostate and preputial gland (P<0.05). But they couldnot influence the weight of seminal gland (P>0.05). After intragastric administration, interstitial cells between glandular cells and glandular epithelium of prostate were shrinked.Interstitial cells between glandular cells were significantly reduced. (2) Sinapine,β-sitosterol and some chemical components of volatile oil could permtate theblood-prostate barrier into the prostate tissue. (3) The extractions also could inhibitangiogenesis on CAM model. (4) The extractions could reduce the expression of VEGFand bFGF (P<0.05), but not TGFβ1 (P>0.05). (5) Sinalbin and sinapine could reduce theexpression of AR (P<0.05), butβ-sitosterol and volatile oil could not reduce it (p>0.05).This paper firstly reported that sinapine,β-sitosterol and some chemical componentsof volatile oil could permtate the blood-prostate barrier into the prostate tissue. Sinalbin,sinapine,β-sitosterol and volatile oil could inhibit angiogenesis on CAM model. Sinalbin, sinapine,β-sitosterol and volatile oil could reduce the expression of VEGF and bFGF.Sinalbin and sinapine could reduce the expression of AR.
