Objective: To analyze the change of Th1, Th2 cells balance and to measure the changes of CD38 expression of CD8~+ T lymphocytes in liver transplant recipients with and without Human Cytomegalovirus(HCMV) disease.Methods:1. Peripheral blood mononuclear cells(PBMCs) were isolated and stimulated by PMA/Ionomycin/Monensin. INF-r and IL-4 production by CD4~+ T cells and CD8~+ T cells in PBMC were analyzed by using dual color fluorescence activated cell sorter(FACS) analysis from liver transplant recipients with versus without HCMV disease.2. CD8~+ T lymphocytes were mixed with corresponding fluorescence labled McAb, CD38~+CD8~+ expression was detected by flow cytometer.Result:1. The average percentages of Thl and Tcl in HCMV disease recipients were significantly lower versus in uninfected recipients(P<0.05).2. The average percentages of Th2 and Tc2 were similar in HCMV disease and uninfected liver recipients(P>0.05).3. The ratios of CD4/CD8 were significantly lower among HCMV disease recipients(P<0.05).4. The CD38 expression of CD8~+ T cells was markedly increased in HCMV disease recipients versus in uninfected recipients(P<0.01).Conclusion:1. Quantitative analysis the CD38 expression of CD8~+ T cells can be used as one of the sensitive and special diagnostic indexes with HCMV infection. 2. Low level of Thl cells seem to correlate with HCMV disease in liver transplantation recipients.
