Objective: To investigate the prevalence of the NAT2 gene in Tianjinpopulation and to compare the distribution of NAT2 genotype betweenthe SLE patients and the health people. The relationship between theNAT2 genotype and SLE also been analysis. Furthermore, Thecontribution of environment exposure factor, such as poison contact, haircolor, and eyebrow tattoos, to the pathogenesis of SLE also beeninvestigated.Methods: A 1:1 matched case-control study was employed in thestudy. 123 SLE patients and 123 healthy controls were enrolled. Thedistribution of M1, M2 and M3 alleles of NAT2 were detected byPCR-RFLP technique. The association between NAT2 polymorphismsand SLE risk was analyzed.Results The prevalence of NAT2 slow acetylator genotypes in Tianjinpopulation was 13.41%. There was no significant association wasobserved between NAT2 acetylator genotypes and SLE risk (P>0.05).The distribution of fast and slow acetylator genotype was significantly different between the SLE cases and controls (x~2=4.235, P=0.04)。Hair color was associate with the increased risk of SLE(OR=0.493,95%CI 0.279-0.871), slow NAT2 acetylator genotype carrier was relatedwith increased risk of SLE, especially in hair color population(OR=0.233; 95%CI 0.063-0.861). eyebrow tattoos also associated withhigh risk of SLE(OR=0.326, 95%CI 0.178-0.598), however, there is nosignificant difference of distribution of fast and slow NAT2 between theSLE and control group(P>0.05) after stratification. Also, there is noassociation between the polymorphism of NAT2 and the injury ofimportant organs OF SLE patient(P>0.05).Conclusion: There is NAT2 polymorphism in the Tianjin population.The distribution of fast and slow acetylation genotype of N-acetyltrans-ferase 2, the history of hair color and eyebrow tattoos were associatedwith the onset of SLE. People with the history of hair color have higherrisk of SLE than the control if they carrying the slow NAT2 gene. Thereis no association between the polymorphism of NAT2 and the injury ofimportant organs of SLE patient.
