Objective: To investigate the expression of RAB5a, MMP-9,ICAM-1and CD44v6 in colorectal cancer, with the expression of tumor-adjacent colorectal carcinoma tissues and normal tissues. And examine the relationships between the expression of RAB5A, MMP-9,ICAM-1and CD44v6 in colorectal cancer and clinicopathologic characteristics, and attempt to seek the best way of diagnosis, evaluating pernicious degree, metastasis in colorectal cancer.Methods: S-P immunohistochemical technique was used to examine the expressions of MMP-9, RAB5a, CD44v6 and ICAM-1 in 46 primary colorectal tumors and tumor-adjacent colorectal carcinoma tissues (3~5cm) and normal tissues. The expressional levels of these adhesion molecules were compared between primary tumors and tumor-adjacent colorectal carcinoma tissues and normal tissues, analyzed with clinical pathology.Results: 1 The positive expression rates of RAB5a protein which was located in cytoplasm and the membrane of the cell were 100%(46/46), more than tumor-adjacent colorectal carcinoma tissues (43.48%, 20/46)and normal tissues(4.35% 1/23),which is obviously diversity compare whit tumor-adjacent colorectal carcinoma tissues and normal tissues (P<0.01,P<0.01).The expression of RAB5a in colorectal cancer was high in the patients with lymph node metastasis ,Duke's stage and differentiation grade(P<0.01,P < 0.05,P < 0.05). The expression of RAB5a has no relationship with gender, age, tumor site, infiltration depth(P > 0.05).2 The positive expression rates of CD44v6 which was located in cytoplasm and the membrane of the cell were 73.91% (34/46), more than tumor-adjacent colorectal carcinoma tissues 36.95% (20/46)and normal tissues4.35%(1/23), which is obviously diversity compare whit tumor-adjacent colorectal carcinoma tissues and normal tissues(P<0.01,P<0.01).The expression of CD44v6 in colorectal cancer was high in the patients with lymph node metastasis ,Duke's stage and differentiation grade, infiltration depth (P < 0.05,P < 0.05,P <0.05,P < 0.01).3 The positive expression rates of MMP-9 which was located in cytoplasm and the membrane of the cell were 73.91% (34/46), more than tumor-adjacent colorectal carcinoma tissues32.96% (17/46)and normal tissues13.04%(3/23),which is obviously diversity compare whit tumor-adjacent colorectal carcinoma tissues and normal tissues(P<0.01,P<0.01).The expression of MMP-9 in colorectal cancer was high in the patients with lymph node metastasis, Duke's stage and differentiation grade(P < 0.05,P < 0.05,P <0.05,P < 0.01). 4 The positive expression rates of ICAM-1 protein which was located in cell membrane and the membrane of the cell were78.26%(36/46), more than tumor-adjacent colorectal carcinoma tissues 43.47%(20/46) and normal tissues 13.04%(3/23) which is obviously diversity compare whit tumor-adjacent colorectal carcinoma tissues and normal tissues(P<0.01,P<0.01).The expression of ICAM-1 in colorectal cancer was high in the patients with lymph node metastasis, Duke's stage and differentiation grade(P<0.05,P<0.05,P <0.05,P < 0.01).5 To apply Spearman rank analysis, 46 cases of the positive expression of RAB5a appear in MMP-9 simultaneously34 cases(r=0.434), 33 cases in ICAM-1(r=0.539), 32 cases in ICAM-1(r=0.346). 32 cases of the positive expression of CD44v6 appear in 27 cases of MMP-9 simultaneously r=0.404), 22 cases in ICAM-1(r=0.527). 34 cases of the positive expression of MMP-9 appear in 28 cases of ICAM-1 simultaneously(r=0.374).Conclusion: The positive expression rates of MMP-9, RAB5a, CD44v6 and ICAM-1 are high than tumor-adjacent colorectal carcinoma tissues and normal tissues, which in colorectal cancer was high in the patients with lymph node metastasis ,Duke's stage and differentiation grade, infiltration depth. In colorectal cancer, bad grade, the positive expression rates of lymph node metastasis, high Duke's stage and low infiltration depth, the positive expression rates of MMP-9, RAB5a, CD44v6 and ICAM-1 are obviously to enhance. To research their characteristic of expression and analyze their relation, we find their expression have intently relationship. The four numerators in the surface of cell examined may guide diagnosis, estimate malignant degree, potential metastasision in colorectal cancer. we can consider them as a combined index in diagnosis and therapy. we can apply the combined therapy for the such patient.
