Objective: This experiment succeeds to make animal model. Through whole studies on inside genital-system damage and hormone level change of pregnant rats, nervous-system injure of fetus rats by chronicity cadmium touch at gestation period, as well as the antagonism and mechanism of different concentr astragalus mambranbceus injection to toxicity of Cd. We identify generate toxicity of Cd further and it is a height doubtful environmental internal-secretion interfering-substance, to explore the effect of astragalus mambranaceus injection. It provides that scientific evidences to illuminate harmful effects by chronicity Cd touch about pregnant women and filial on animal studies and that theory these to prevent and cure of cadmium toxicity. Meanwhile this develop a new pathway for searching effective treatment prescription of cadmium toxicity on traditional and western medicine.Method:1 According to 2:1 ratio mating after 50 femal and 25 male SD rats were adapted a week. The day of finding pessulum was first-day of conception.40 conceived SD rats were divided into 4 groups randomly: low concentration astragalus mambranaceus+cadmium group(A1), high concentration astragalus mambranaceus+cadmium group(A2), cadmium group(Cd) and control group(C). Cadmium water(40mg/L) was given by drinking to A1,A2 and Cd groups everyday. Distilled water was given at the same dosage by drinking to C group. Astragalus mambranaceus injection 5g/kg/d or 10g/kg/d were given by intraperitoneal injection to A1 and A2 groups everyday. Distilled water was given at the same dosage by intraperitoneal injection to Cd and C groups. All groups were given common forage everyday.2 Pregnant rats were anesthed by pentobarbital(40mg/kg) at the 20th day, and obtained blood form the hearts and centrifuged blood-serum. The contents of cadmium(Cd) ,Estrogens(E2), Progestogen(P) and Prolactin(PRL) were evaluated. The activity of Glutathione peroxidase(GSH-Px) and Hemocuprein(SOD) were measured. Meanwhile, placenta was taken out and evaluated the content of Cd, as well as it was observed by electron microscope. Subsequently, embryos were taken out quickly, weighed accurately and measured body and tail length. Their lives and brains were acquired quickly at the ice disc and the conent of Cd was evaluated. Another, their brains were evaluated the content of Acetylcholine(Ach).Result:1 Two pregnant rats of only Cd group appeared general toxicosic representation within the epression, anorexia, clothing hair glossiness and diarrheal.2 Contrast to C group, the contents of blood-serum and placenta cadmium increased in pregnant rats of Cd group. Contrast to Cd group, the contents of blood-serum and placenta cadmium degraded in A1 and A2 groups, but there were not obviously difference between A1 and A2 groups. They existed obviously correlation between the contents of blood-serum and placenta cadmium, and not only they were regression but also correlate relation. It showed that its of placenta cadmium increased following the content of blood-serum cadmium.Contrast to C group, the activity of GSH-Px and SOD enzyme degraded obviously in the blood-serum of Cd group. Contrast to Cd group, the activity of two enzyme advanced in A1 and A2 groups. Futher, the activity of two enzyme advanced obviously in A2 group, and it existed dosage-reaction effect.Contrast to C group, the level of E2 and P degraded obviously in the blood-serum of Cd group. Contrast to Cd group, the level of E2 and P advanced obviously in A2 group, but the level of them did not change in A2 group, and difference was significance between A1and A2 group. It existed dosage-reaction effect. The level of PRL did not change in the blood-serum of all groups, and difference was not significance in all groups. 3 The result showed of placenta morphology(1)Light microscope showed: Contrast to C group, Cd group placenta of pregnant rats existed villi interstitial edema, microthrombus of basilar and labyrinthus part, large area necrosis and array disord of villis constit. Structure process of astragalus mambranaceus injection group lighten. There is leukomonocyte and plasmocyte infiltrate,and endochylema puff in the A1 group. There is merely lighten patho change, it neared basically C group, and only there is few leukomonocyte infiltrate.(2)Electron microscope showed; Contrast to C group, Cd group placenta of pregnant rats existed few microvilli at syneytiotrophoblast surface, caritation and pyknosis of chondriosome, and rough endoplasmic reticulum degranulated ultramicrostructure of placenta damage lightly in A1 group. A2 group neared basically C group. The ultramicrostructure of placenta was destroyed severity in Cd group and was destroyed slightly in A1 group of the pregnant rats. The ultramicrostructure of placenta was normal fundamentally in A2 group.4 The indicatrix that growth and development about pregnant rats(body weight and body-tail length).Body-tail length of did not exist significant deviation in all groups. but contrast to C group, there existed downtrend in Cd group . The body weight lighten obviously in Cd group, and the difference existed significance. 5 Contrast to C group, cadmium content of liver about fetus rats increased obviously in Cd group, and those of brain was also to increase, but not as good as liver. Contrast to Cd group, cadmium content of liver and brain degraded obviously in A2 group, but those of liver and brain did not change in A1 group, and between A1 and A2 group the difference existed significance. That showed dosage-reaction effect.6 Contrast to C group, content of Ach of brain tissue about fetus rats degraded obviously in Cd group. Contrast to Cd group, those increased obviously in A2 group, but those did not change in A1 group. And between A1 and A2 group difference existed significance. That showed dosage-raction effect.Conclusion:1 Astragalus mambranbceus injection can rivalry the toxicity of cadmium about pregnant rats and fetus rats. The mechanism of action possibly related to astragalus mambranbceus injection clearing oxygen free radical.2 That leaded to diff-extent injure to genesis endocrine system of pregnant rats after touching chronicity cadmium, and texture and function of placenta changed, leading to changing obviously of hormone. Further, it identified that reproductive organ of pregnant rats became target organ of cadmium, and cadmium was a height doubtful environmental internal-secretion interfering-substance.3 Cadmium can enter internal body of fetus rats by placental barrier, it influenced the growth and development of fetus rats. Cadmium poisoned the nervous system of fetus rats by blood brain barrier.
