Objective:To study the expression of COX-2 and PCNA in meningioma and the relationship between COX-2 and the grade. To investigate the function of COX-2 and PCNA in meningioma. To investigate the value of COX-2 in clinical treatment of meningioma.Material and method: 42 samples of patients,who underwent operations at the second Affiliated Hospital , Hebei Medical University from 2005 to 2006, and 5 samples of normal brain tissue were collected. Chemiotherapy, radiotherapy and immunosuppressive therapy were not performed before operation. All the samples were confirmed by histopathological diagnosis.Immunohistochemical stains(SP) were performed to examine the expression of COX-2 in all samples. The expression of COX-2mRNA were checked by in situ hybridization technique. Immunohistochemical stains(PV) were performed to examine the expression of PCNA.Applying SPSS11.5 statistical package, carry out chi-square test, multi-sample mean multiple comparison and correlation analysis on results. P value less than 0.05 was considered statistically significant. Results: There were not positive expression of COX-2 in the normal brain tissue.In 30 cases of meningioma in gradeâ… , the positive expression of COX-2 were 8 cases(26.7%); in 6 cases of gradeâ…¡, the positive expression of COX-2 were 3 cases(50%); in 6 cases of gradeâ…¢, the positive expression of COX-2 were 5 cases(83.4%). With the grade of meningioma higher, there were more cases which were positive expression of COX-2(P<0.05). In the gradeâ…¡andâ…¢meningioma, it was no evidence that there were more cases which were the positive expression of COX-2 with the grade of meningioma higher (P>0.05). But between the gradeâ… meningioma with the gradeâ…¡andâ…¢, it was evidence that there were more cases which were the positive expression of COX-2 with the grade of meningioma higher(P<0.05).By in situ hybridization, there were not positive expression of COX-2mRNA in the normal brain tissue. In addition, there were higher positive expression of COX-2mRNA in the high grade meningioma than the low grade meningioma and normal human brain tissue. The positive expression rates of COX-2mRNA in the normal brain tissue and gradeâ… ,â…¡,â…¢Meningioma were 0%,30%,66.7% and 83.4%. With the grade of meningioma higher, there were more cases which were positive expression of COX-2mRNA(P<0.05). In the gradeâ…¡andâ…¢meningioma, it was no evidence that there were more cases which were the positive expression of COX-2mRNA with the grade higher (P>0.05). But between the gradeâ… meningioma with the gradeâ…¡andâ…¢, it was evidence that there were more cases which were the positive expression of COX-2mRNA with the grade higher(P<0.05).PCNA were expressed typically in cell karyon. There were the lowest expression in normal human brain compared with every grade of meningioma with a significant difference (P<0.01). Expression of PCNA in gradeâ…¢meningioma were higher than gradeâ…¡and gradeâ… meningioma with a significant difference (P<0.01). Expression of PCNA in gradeâ… meningioma were lower than gradeâ…¡meningioma with a significant difference (P<0.01).Used correlation test. Statistical analysis of results indicated COX-2 expression was correlated with PCNA expression in meningioma(r=0.828, P<0.01).Conclusions:1 The expression of COX-2 and COX-2mRNA in the meningioma were obviously more than them in the normal brain tissue, and the expression of COX-2 and COX-2mRNA in the high grade meningioma were obviously more than them in the low grade meningioma. It shows that the activation of COX-2 was correlated to the degree of the multiplication in meningioma.2 The expression of PCNA in the meningioma were more and more with the grade higher and higher. It is illuminated that the cells of increment phase in the high grade meningioma were obviously more than them in low grade meningioma and normal brain tissue.3 With the degree of the expression of PCNA was high,the expression of COX-2 corresponded to high. The correlation between them was statistically significant. It indicates that the cells of increment phase in meningioma samples of high expression of COX-2 were obviously more than them in meningioma samples of low expression of COX-2.
