Objective:Making disuse osteoporosis model of rat by denervating method,identify method of denervating to produce osteoporosis.researching relationship of neurotransmitter SP with transcription factor OPG and RANKL in disuse osteoporosis of rat,research mechanism of disuse osteoorosis.Material and methods:purchase 40 maleness rat of Wistar,random distribute two groups:first group,experiment group(model group,sciatic nerve break group,CON),To cutting sciatic nerve 1.00cm in right post crura of rat for disuse osteoporosis.two group:normal control(sham operation,SHAM):Progress sham operation to right post crura of rat.After 6 week,12 week,detect every group right post crura of rat femoral bone density(BMD),bone tissue pathematology detection,using immunity hixtochemistry method detect femoral SP and transcription factor OPG and RANKL expression,Use IDA-2000 figure pattern analytical system to analysis image.Result:1,Aftet 6 and 12 week of operation,experimental group and normal control femoral BMD in right post crura of rat markly less than normal control(P<0.05)have significance statistics.control group:bone trabecula alignment closely.experimental group:bone trabecula conspicuous decrease,construction alignment disorder,rarefaction,formatio reticularis absence,bone trabecula thickness thinningz,blank widen.2,After 6 weeek of operation,experimental group rat right post crura femoral substance P immunity histochemical expression markly less than normal control(P<0.05),having significance of statistics;But after 12 week of operation,SP have not deference with normal control(P>0.05) having not significance of statistics.3,Result of OPG and RANKL expression in experimental group and normal control displayl: masculine expression of OPG and RANKL locate kytoplasm,buffy,expressing in osteoblast.After 6 weeks,OPG expression become powerful masculine in experiment group,but become puny masculine in control group.Two groups have markly difference(P<0.05).After 12 week,expression of OPG become masculine in control group,also become masculine in experiment group.Two groups markly have not deference(P>0.05).After 6 week,RANKL expression become puny masculine in control group,but become powerful masculine,two groups markly have difference(P<0.05).After 12 week,RANKL expression become masculine in control group,also become masculine in experiment group,two groups markly have not deference(P>0.05).Conclution:1,appling denervation method to produce disuse osteoporosis model is feasible.2,After denervation,SP content of femoral bone decrease.3,SP become positive correlation with OPG,and negative correlation with RANKL.4,After denervation,SPcan induce osteoporosis through changing expression of OPG and RANKL.
