Objetive:To investigate the frequency of the CD4+CD25+ regulatory T cells(Treg)in the patients with ovarian carcinoma,and to study the correlation between CD4+CD25+ regulatory T cells and clinicopathological characteristics of patients with ovarian carcinoma;To explore the correlation between CD4+CD25+ Treg cells and different cytokines.Methods:The percentages of CD4+CD25+ Treg,CD44 and the levels of interleukin 2(IL-2),tumor necrosis factor-α(TNF-α),interferon-gamma(INF-γ),interleukin 4(IL-4),interleukin 6(IL-6),interleukin 10(IL-10)in the peripheral blood lymphocytes from 53 patients With ovarian carcinoma,44 patients with benign diseases,45 healthy people were determined by flow cytometry.The levels of CA199,CA125 were detection by ELISA.Results:(一):the proportion of CD4+CD25+Treg(21.95±6.80)%,CD44(128.45±34.72)%in the peripheral blood lymphocytes from patients with ovarian carcinoma were significantly increased compared with those from benign diseases(12.57±5.12)%,(83.75±19.49)%,and the control(10.77±3.02) %,(79.92±21.83)%,(p<0.001);the proportion ofCD4+CD25+ Treg(21.95±6.80)%,CD44(128.45±34.72)%in the properative patients were higher significantly than those from after operative patients(16.41±5.84)%,(103.41±37.25)%,(p<0.001)。(二):CD4+CD25+ Treg,CD44 were not related to age and profession(P>0.05).There was a significant increase of CD4+CD25+ Treg(23.58±7.08)%in serous carcinomas compared with those from mixed tumors(18.30±5.46)%,(p<0.05),but did not demonstrate a difference compared with those from mucinous carcinomas(22.06±6.08)%;CD4+CD25+ Treg(28.79±4.26)%,CD44 (157.24±33.57)%inⅣstage showed more rapid increase thanⅢstage(20.01±6.27)%,(124.31±29.46)%,andⅠ-Ⅱstage(17.97±4.50)%,(116.08±30.93)%,(p<0.001);The percentage of CD4+CD25+ Treg(23.61±6.52)%,CD44(143.38±35.11)%in invasive patients,(24.05±6.74)%,(146.32±35.87)%in lymph node patients,(24.06±6.73) %,(146.52±35.86)%in ascites patients were higher than non- invasive patients(18.96±3.33) %,(108.95±26.02)%,non-lymph node patients(19.73±3.51)%,(115.86±23.71)%,non-ascites patients(19.70,3.49)%,(115.17±22.67)%.(p<0.05-p<0.001).There were more increasing of the proportion of CD4+CD25+(25.05±5.96)%,CD44(152.50±32.49) %in poor-differentiated patients than those from well- differentiated patients(16.03±0.80)%,(105.95±15.43)%,(p<0.05).(三):The concentration of IL-2(0.87±0.25)pg/ml,TNF-a(1.09±0.34)pg/ml,IFN-r(7.56±2.25)pg/ml in the patients with ovarian carcinoma were lower than those from healthy people(2.38±1.09)pg/ml,(3.28±1.36)pg/ml,(19.04±4.59)pg/ml,(p<0.001).but the lever oflL-6(86.28±55.92)pg/ml was striking increase in the patients with ovarian carcinoma compared with that in healthy people(5.04±2.69)pg/ml, (p<0.001).no differences were detected in IL-4,IL-10.By nonparametric correlation,The percentages of CD4+CD25+ Treg has a negative correction with concentration of IL-2 (r=-0.658,p<0.05),TNF-α(r=-0.591,p<0.05),INF-γ(r=-0.695,p<0.05),and has a positive correction with level of IL-6(r=0.784,p<0.001).(四):The proportion ofCD4+CD25+ Treg has a positive correct;on with CD44(r=0.406,p<0.05),CA125;(r=0.563,p<0.001),But no correction with CA199(r=0.036,P>0.05).Conclusion:(一):There are increasing of the proportion of CD4+CD25+ Treg,CD44 in the patients with ovarian carcinoma,which is probably an important reason of reduced immunological function of ovarian carcinoma.(二): the proportion of CD4+CD25+ Treg,CD44 were closely related to FIGO,types of histology,degree of differentiation,tumor invasive,lymph node status,form of ascites.(三):The proportion of CD4+CD25+ Treg has a negative correction with IL-2,TNF-α,INF-γ;a positive correction with IL-6.The cytokines may have some effects in the immune suppression mechanism of CD4+CD25+ regulatory T cells.(四):Detection of CA125,CD44 and some cytokines related to the cellular immunological function of ovarian carcinoma at the same time have important significance to control and treat ovarian carcinoma.
