Objective: To study the role of E-selection and macrophageinflammatory protein-2 (MIP-2)in the airway inflammation in a rat modelof chronic obstructive pulmonary disease(COPD).Methods: Twenty-four male SD rats were randomly divided into anormal control group and a COPD model group. The rat model ofCOPD was established by exposure to cigarette smoking. Rats' lungfunction,pathologic features of lung tissues and inflammatory celldifferentials in bronchoalveolar lavage fluid(BALF) were investigated.Immunohistochemistry was carried out to examine the expression ofE-selection in lung tissue.The levels of MIP-2 in BALF, serum andlung tissues were measured with ELISE.Results: (1)lung function tests showed that FEV0.3,FEV0.3/FVCand PEF values were significantly lower in the group with COPD than inthe normal control group(all P<0.05).(2)Pathological change of the modelgroup consistents with that of COPD. MLI in the COPD model groupwas higher than that in the normal control group while MAN wasdecreased in the COPD model group compared with the normalcontrol group (all P<0.05). Total leukocyte counts ,neutrophil countsand neutrophil proportion were signicantly increased in the COPD modelgroup as compared with the normal control group (all P<0.05). (3)The expression of E-selection on bronchopulmonary vein endothelial cells ofthe COPD model group significantly increased as Compared with thenormal control group(P<0.05) and was positively correlated with theneutrophil counts in BALF in the COPD model group(r=0.809,P<0.01).(4)The levels of MIP-2 in BALF and lung tissues in the rats with COPDwere signicantly higher than those in the rats of normal and were bothpositively correlated with the neutrophil counts in BALF in the COPDmodel group(r1=0.893,P1<0.01; r2=0.716, P2<0.05).Conclusion: (1)The rat model of COPD which was established by theway of giving passive smoking for eighty days was successful,which wasproofed by lung function tests and pathological observation. (2) BothE-selection and MIP-2 may be involved in the process of airwayinflammation in COPD.
