Objectives: To investigate the expressions of Oct4 and PSCA of ECC, and to search for the evidence in support of the stem cell theory of carcinogenesis of ECC.Methods: Expressions of Oct4 and PSCA were detected in the 43 tissues of ECC as well as 10 normal tissues of bile duct by immuno-histochemistry (EnVisionTM). The different expression levels in two kinds of specimens were dealed with statistical analysis, so did the correlation between the expression and the clinic pathologic character.Results:1. The positive rates of Oct4 and PSCA in the ECC group were 44.2 % and 46.5%. They were significantly higher than that in normal group(0,10%)(P<0.05).2. The expressions of Oct4 were associated with differentiation and invasion (P<0.05), but not with the sex,age of patients and the size, location of tumors(P >0.05).3. The expression of PSCA had no relationgship with the sex, age of patients and the differentiation ,size,location of tumors(P >0.05).Conclusions:The significant differences between expressions of Oct4 and PSCA implied that there were "stem-like cells" existing in the tissue of ECC. Expression of Oct4 was related to the differentiation and invasion of tumor, and ti might be an important index for evaluating the prognosis of ECC.
