Objectives: To observe the osteoblastic-like cells' shape after different continuously compressive pressure, and investigate the effect of continuously compressive pressure on the expression of integrinα5 and integrinβ1mRNA in osteoblastic-like cells in vitro。Methods: 1.To resuscitate MG-63 cell strain, and culture the cells。2.cells were cultured in the utensil, divided into two groups: control group and pressure group; then divided the pressure group into three groups: 15kpa,30kpa,45kpa。There are 15 samples in every group, using 3 samples in every pressure experiment. The pressure duration of each group is 1hr,4hr,8hr,12hr,24hr separately。And collected the cells after treat each sample, then measured the expression of integrin a5 and integrinβ1mRNA。Results: 1. The morphology of osteoblastic-like cells had no significant change after cultured in the mechanical pressure model。2. The expression of Osteoblastic-like cells'integrinα5,β1 mRNA enhanced with time when treated with 15kpa and 30kpa, had significant difference with control group and 45kpa grope, theα5,β1 mRNA expressed highest when treated 30kpa after 24hr。The expression ofα5,β1 mRNA enhanced when treated 45kpa after 1hr and 4hr, but reduced with time。Conclusions: 1.We can use this kind of pressure model to do continuously compressive pressure experiment。2.The physiological range of pressure stimulates the expression of integrin a5,β1 mRNA in osteoblastic-like cells, when the pressure force overloaded, the expression will reduced with time。...
