Objective: to observe effect of acidic fibroblast growth factor on proliferation and dose-effect relationship of tendon cells in vitro.Methods: The 3th passage tendon cells of galactic rabbit in vitro were planted in 60 wells of a 96- well culture plate and every well contained 4×10~3 cells. They were divided into 12 groups and every group contained 5 wells. According to aFGF concentration, no aFGF group, 0.5ng/ml aFGF group, 1.0ng/ml aFGF group, 2.0ng/ml aFGF group, 5.0ng/ml aFGF group, 10ng/ml aFGF group, 20ng/ml aFGF group, 30ng/ml aFGF group, 40ng/ml aFGF group, 50ng/ml aFGF, 100ng/ml aFGF and 200ng/ml aFGF group were named respectively. When all groups being in exponential phase of growth under microscope, they were dyed by MTT method and OD value of every well was detected by enzyme linked immunodetection meter.Results: By contrast to OD mean of control group: 1.0ng/ml aFGF group, 2.0ng/ml aFGF group, 5.0ng/ml aFGF group, 10ng/ml aFGF group, 20ng/ml aFGF group, 30ng/ml aFGF group, 40ng/ml aFGF group, 50ng/ml aFGF group, 100ng/ml aFGF group, 200ng/ml aFGF group had significant difference(P<0.05) for promoting proliferation of tendon cells; 0.5ng/ml aFGF group has no significant difference (P>0.05) for promoting proliferation of tendon cells; When aFGF concentration varying from 0.5ng/ml to 20ng/ml, OD mean of every group increased gradually, while from 20ng/ml to 200ng/ml, it decreased step by step.Conclusions: 1.0,2.0,5.0,10,20,30,40,50,100,200ng/ml aFGF could promote proliferation of tendon cells; 0.5ng/ml aFGF has no significant effect on proliferation of tendon cells; aFGF concentration varying from 0ng/ml to 200ng/ml, proliferation ability of tendon cells in every group firstly increased(1ng/ml~20ng/mlaFGF) gradually, then decreased (20ng/ml~200ng/mlaFGF)step by step.
