Objective: The aim of this study was to observe the proliferationability of the rabbit NP cells that cocultured with MSCs to get a way forharvesting more NP-like cells in vitro.Methods: The BMSCs and NP cells were got from the New ZealandRabbit, and they were cocultured with 10% calf serum. According to thecell ratio between BMSCs and NP cells, the cells were divided into sevengroups: group A was NP cells control group; group B was BMSCscontrol group; the ratio between NP cells and BMSCs cells in group Cwas3:1, the ratio of group D was 2:1; group E was 1:1; group F was 1:2;group G was 1:3. The cell population was counted and theimmunohistochemical method was used for comparing the proliferationability of the NP cells.Results: The population of cells in group A was significant lowerthan the other groups (P<0.01); group B was significant lower thangroup C to group G(P<0.01); group C was significant higher than groupE to group G (P<0.01), but there was no significant difference betweengroup C and group D (P>0.05); group E was significant higher thangroup F and group G (P<0.01); group F was higher than group G (P<0.01). In group A, all the cells were colored and there was no positivecells in group B. The positive cells and negative cells were coexisted in group C to group G. The increase of positive cells in group C was higherthan that in group E to group G (P<0.01), but there was no significantdifference between group C and group G (P>0.05); group E was higherthan group F to group G (P<0.01); group F was higher than group G (P<0.01).Conclusions: The coculture system that rabbit BMSCs wereconnected with NP cells could promote the proliferation of NP-like cells.The proliferation ability of NP-like cells increased with the ratio betweenNP cells and BMSCs when the ratio was between 1:3 and 2:1; there wasno significant association between proliferation ability of NP-like cellsand the ratio when the ratio between NP cells and BMSCs was 2:1 and3:1.
