Study On The Influence Of Hyperbaric Oxygen To Nasopharyngeal Carcinoma CNE-2 Cell Line's Radiotherapy

[Objective] The purpose of this research was to investigate effect of radiotherapy for nasopharyngeal carcinoma cells after hyperbaric oxygenation therapy, and to approach the influence of hyperbaric oxygen to the radiotherapy of nasopharyngeal carcinoma in cellular level.[Methods] The nasopharyngeal carcinoma CNE-2 cells were distributed randomly to A,B,C and D four groups. A group was radiated 2Gy rather after 0.22MPa hyperbaric oxygenation, and the duration of hyperbaric oxygenation was 60 min. B group was only treated with 0.22MPa 60 min's hyperbaric oxygen. C group was radiated 2Gy only. And D group was treated blankly as control group. The influence of growth inhibiting of the nasopharyngeal carcinoma CNE-2 cells had been investigated by MTT method. The condition of cell death had been observed under PI staining. We analyzed the change of the cell apoptosis and cell cycle by the flow cytometry (FCM). And the cell pathological change had been viewed under electron microscope.[Result] The A,B and C groups cells' growth were inhibited and A's growth inhibiting rate was highest, C was the second and B was the third. There were statistics differences among the there groups. The death rate and apoptosis rate of A group cells were the highest in the four groups (P<0.05). Compared with D group, the death rate of the B group cells was higher (P<0.05), and apoptosis rate was higher, but there is no statistics difference on apoptosis rate (P=0.142). The death rate and apoptosis rate of the C group cells were higher than the D group's, but were lower than A group's. The cell cycle was changed after hyperbaric Oxygenation. After hyperbaric oxygenation the rates of cell stages G2 and G2/G1, which were sensitive to radiotherapy, were higher than non-HBO groups; and the rate of cell stage G1, which was insensitivity to radiotherapy, was lower than non-HBO groups. And in hyperbaric oxygen therapy groups, the rate of cell stage S was higher than in non-HBO groups.[Conculsion]①Hyperbaric oxygenation could sensitize the radiotherapy effect of nasopharyngeal carcinoma CNE-2 cells.②Treated with hyperbaric oxygen only, the growth of nasopharyngeal carcinoma CNE-2 cells were inhibited and apoptosis was increased.③Hyperbaric oxygenation could influence the cell cycle of nasopharyngeal carcinoma CNE-2 cells.
