Expression and clinical significance of fatty acid synthase in ovarian cancerobjectiveOvarian cancer is a common malignancy of female genital mutilation, the incidence of it ranks the third. But died of ovarian cancer, which accounts for the top types of gynecologic tumors. So far, statistics on domestic and clinical data, the five-year survival rate is only 25-30%,so the early diagnosis target of ovarian cancer is important for improving the survival rate of patients. The malignant behavior of tumor tissue is closely related with the special substance metabolism and energy metabolism. fatty acids plays an important role in the Energy metabolism and structure function of cells. Fatty acid synthase (FAS) is a key enzyme in the synthesis of endogenous fatty acids. So ,in recent years the expression of the enzyme in cancer has become a focus research. At present, some research shows that FAS is highly expressed in breast cancer, prostate cancer, lung cancer and other malignant tumors, which is associated with poor prognosis. However, the study of FAS in ovarian cancer is seldom, the purpose of this study is to discuss the expression in ovarian cancer and its clinical significance by methods of immunohistochemistry and RT-PCRMethods1. patients and specimens:10 cases of benign ovarian . 10 cases of borderline tumors, 30 cases of malignant tumors 3 carcinomas were grade 1,9 grade 2,and 18 grade 3;2 cases ofⅠ, 5 cases ofⅡ, 21 cases ofⅢ,2 cases ofⅣ). 10 cases of normal ovarian. 2. To detect the expression of FAS protein in different ovarian tissue by immunohistochemistry PV-9000.and analysis the relationship between FAS and ovarian cancer.3. To detect the expression of FAS mRNA in different ovarian tissue by RT-PCR,and analysis the relationship between FAS and ovarian cancer.4. Statistical data analysis : using SPSS 11.0 software. For immunohistochemistry method of analysis using chi-square test and Fisher′s exact probability test; RT-PCR using the unpaired Student′s t test. The difference was significant as to 0.05Results1. In normal ovarian tissue, benign tumors, borderline tumors and malignant tumors, the positive rate of FAS protein was 0/10,2/10 ,5/10,90%. After statistical analysis, The difference between normal and borderline group,normal and malignant group, benign and malignant group,borderline and malignant group have statistically significance (P<0.05); the difference between normal and benign group. Borderline and benign group have not statistically significance(P>0.05).2. There was no significant difference between FAS and degree of differentiation and lymphatic metastasis(P>0.05),but FAS correlated with clinical staging. The difference was significant (P<0.05).3. In normal ovarian tissue, benign tumors, borderline tumors and malignant tumors, the expression of FAS mRNA increased gradually, and malignant group was significantly higher than the normal group. After statistical analysis, we found that the difference between normal and borderline group ,normal and malignant group, benign and borderline group ,benign and malignant group, borderline and malignant group were statistically significant(P<0.05).4. In ovarian cancer, there was no significant difference between FAS mRNA and degree of differentiation and lymphatic metastasis(P>0.05),but FAS mRNA correlated with clinical staging. The difference was significant (P<0.05). ConclusionsThe expression of FAS is related with malignant degree of ovarian tumors. FAS can be used as indicators of early diagnosis of ovarian cancer, and may provide a new evidence for early diagnosis of ovarian cancer...
