ObjectiveAnalyzing the differences in protein profile between endometrial carcinoma andnormal endometrium using the two-dimensional electrophoresis (2-DE) and massspectrometry of proteomics, to search proteins expressed differentially in humanendometrial carcinoma.Methods1. Separated the different proteins of endometrial carcinoma and normalendometfium by two-dimension gelelectrophoresis.2. Analyze the reproducibility of gels of endometrial carcinoma and normalendometfium, and the differential proteins by ImageMaster 2D platinum.3. Identified the differential proteins by mass spectrometry and look into theprotein database online.4. Validated a few of the identified proteins by RT-PCR.Results1. The images of endometrial carcinoma and normal endometrium were analyzedsuccessfully and good reproducibility were obtained.2. Analyzing by Imagemaster 2DE software, twenty-seven proteins were founddifferentially expressed in tissue from endometrial carcinoma and normal endometrium.Twenty-two proteins were up-regulated in endometrial carcinoma, five proteins weredown-regulated in endometrial carcinoma.3. After analyzing by MALDI-TOF-MS, fifteen differentially expressed proteinswere identified. P48, ENO1 protein, ATP synthase, Annexin, Elongation factor 1-alpha1, Unknown (protein for IMAGE: 3897065), Peroxiredoxin-6, Human MacrophageCapping Protein Cap G, pyrophosphatase 1, Human Manganese Superoxide Dismutase Mutant Q143n, protein disulfide isomerase-associated 3 precursor, pyruvate kinaseisozymes M1/M2, leucine aminopeptidase(LAP) were down-regulated in endometrialcarcinoma, and Stathmin 1 and alpha-1 antitrypsin were up-regulated in endometrialcarcinoma.4. The mRNA expression of P48, ENO1 protein, ATP synthase, Annexin,Elongation factor 1-alpha 1 increases in the endometfial carcinoma, Stathmin 1decreases in the endometrial carcinoma.Conclusion1. Proteins which expressed differently in endometrial carcinoma may beassociated with energy metabolism, ant-oxidation, cell growth, proliferation,carcinogenesis, signal transduction regulation, molecular chaperones and so on.2. The differential proteins in endometrial carcinoma may provide further insightinto the useful clues for the pathogenesis of endometfial carcinoma.
