PrefaceGlioma is the most common primary neoplasm of central nervous system (CNS) in human beings .Because of its high incidence high recurrence, poor prognosis , there is no radical treatment method up to now .just as other tumors ,the real reason about the occurring of glioma is not clear. it is considered to be related with circumstance ,heredity and dysfunction of immunology. In the present study about this disease ,the relations between the glioma and cytokines are getting more and more attentions .TNF (tumor necrosis factor) is a very important kind of cytokine which has many biological functions, and it makes many important roles in tumor growth, proliferation, differentiation ,inducing cell-death, killing tumor cells and activating immune system .The biological properties of TNF are signaled through its cell surface receptors—TNFR (tumor necrosis factor receptor) .there are many disputes about whether the tumor can produce TNF. At present, it is thought that Central Nervous Systeme(CNS) is not immuno-exempt . The cytotoxicity of TNFαis mainly mediated by TNFR1 .at pathological state, there are humeral immunity and cell immunity still. TNFαcan be produced not only in peripheral circulation, but also by small glioma cell, astrocyte, nerve cell and vascular endothelial cell. Though there are many researches in laboratory about using TNF to therapy glioma ,the relation between TNF and glioma is not clear. We determine the protein expression of TNFαin glioma and surrouding tissue by immunohistochemical methods. This research can help us know the expression of TNFαin glioma of different pathological grades. It provides theory dependence for laboratory research of TNFαmediated glioma cell apoptosis, determining pathologic grade of glioma and clinical antineoplastic treatment.ObjectiveTo evaluation the expression of tumor necrosis factor( TNFα) in glioma and surrounding tissues of tumors and the relationship among the expression of TNFαin the three different clinical pathological gradation in human.MethodsApplying SABC immunohistochemical technique . Examination and statistical research on the expression in 35 cases of human glioma and 8 cases of tumor-around tissue .The 35cases of glioma are graded according to WHO 1993 criteria.ResultsTNFαmainly expressed in cytoplasm , the positive expression rate in surrounding tissue of tumor is 12.50% . the positive rate of TNFαexpression in glioma is 54.29% ,and there is obvious difference in the expression of TNFαbetween glioma and surrounding tissue of tumor (p<0.05) .the positive rate of TNFαexpression in I~II-grade glioma is 73.19%. the positive rate of TNFαexpression in Ill-grade glioma is 25.00% . and there is obvious difference in the expression of TNFαbetween the tumor pathological gradation(p<0.05). the expression of TNFαis correlated with the tumor pathological gradation.Conclusions1,glioma can express TNFα.2,TNFαexpresses higher in glioma than that in surrounding tissue of tumor.3,The expression of TNFαin I~II-grade glioma of human beings is high, in III-grade glioma is low. there is statistic meaning.
