ObjectiveDiabetic cystopathy is one chronicity complication of diabetes. there may be have 40~100 percent incidence in patients with diabetes. The DCP have gravely affected patients' quality of life. It is getting increasing attention from doctor of department of urology, endocrine secretion and department of neurology. Because the early symptom of DCP is very delitescence, only after the detrusor muscle appeared decomp, DCP can be clearly diagnose. It is too late to treat. So it is very necessary to research thoroughly the pathogenesis of DCP. There are many variety of inducement, as we know, such as blood vessel dysfunction, muscle dysfunction and nerves injury, et al. In these inducement , nitric oxide paly a very important role .But there is no dependabily study about the relation of DCP and nitric oxide . in this study ,we determine the contents of nitric oxide and activity of nitric oxide synthase in the bladder 8weeks after diabetes inducted by treptozocin. To evaluate the leveles of nitric oxide and nitric oxide synthase in the bladder 8weeks after diabetes induction. Nitric oxide will been implicated playing a important role in the pathogenesis of diabetic cystopathy.Methods60 healthy male Sprague-Dawley rats were used in the study . The rats were divided into three groups. Group 1(n=20)was the control; group 2(n=10) drank 5% sucrose, replacing water as a control of diuresis; and group3(n=30) received a single intraperitoneal injection of streptozotocin(STZ;50mg/kg), after over night fasting , to induce diabetes (blood glucose level 16.7mmol or greater). The body weight and blood glucose levels were measured weekly . Eight weeks later, the bladders were harvested for studies. After hemagglutinin-esterase stain observation. We measured the levels of nitric oxide,nitric oxide synthase and inducible nitric oxide synthase In the bladders. Methods are nitrate reductase method and colorimetric method.ResultsThe contrast of blood glucose levels is very adversely , with 28.4±8.4 , 6.9±2.8, and 4.8±2.3. we observed that the detrusor of bladder were emerging confused alinement and lamellar necrosis. The levels of nitric oxide were statistically significantly.Increased in the diabetic bladder homogenates compared with those of the other groups(P =0.006,diabetic versus control). The nitric oxide synthase activity were statistically significantly enhanced in the diabetic bladder homogenates compared with those of the other groups(P =0.0027,diabetic versus control). But numerical data contrast between the two control groups is not statistically significantly(P>0.05).Conclusions1,The STZ-induced diabetic rat is a well-established model for diabetes studies.2,Our findings showed that the level of nitric oxide increased in the bladders of the STZ-diabetic rats, the activity of nitric oxide synthas and inducible nitric oxide synthas enhanced in the bladders of the STZ-diabetic rats.3,The increase in the bladders of the STZ-diabetic rats was not mediated by single diuresis.4,The increase of nitric oxide is because that nitric oxide synthase activity was enhanced when pathologic status, especially inducible nitric oxide synthas.5,Nitric Oxide plays a very important role in pathogenesy of diabetic cystopathy.
