Objective 7KC is investigated the influence on NF-κB and whether NF-κB was involved in the effection of J774A.1 cells apoptosis induced by 7KC. Methods J774A.1 cells cultured in vitro were exposed to 7KC.The translocation of NF-κB and the degradation of IκBαinduced by 7KC on J774A.1 cells were detected by immuocytochemisty staining and Western Blot.Histopathological changes were evaluated by hematoxylin and eosin staining.Investigation the effects of PDTC on macrophage apoptosis by 7KC were determined by trypanblue staining and MTT and AnnexinV/Propidium Iodide double-labelled flow cytometry.Results Results of immunocytochemical staining of NF-κB/p65 showed that only cytosol was positively stained in nature cell,and nuclei became positively stained after 7KC stimulation. Results of Western Blot showed that the degradation of IκBαinduced by 7KC in a concentration-and-time-dependent manner.7KC inhibited increase and induced apoptosis and death in J774A.1 cells.PDTC can protective J774A.1 cells from 7KC induced-apoptosis though activating NF-κB.conclution (1)7KC induced activation of NF-κB in J774A.1 cells.(2) 7KC induced activation of NF-κB was mediated by IκBαdegradation.(3) Macrophages apoptosis were induced by 7KC .(4) 7KC-induced macrophages apoptosis was mediated by NF-κB.
