Objective: To study the situation of the cytotoxic activity of CIK cells induced by dendriccells pulsed with antigen on the gastric carcinoma xenografts modified by newcastle dieasevirus.Methods: Mononuclear cells were isolated from peripheral blood of the healthy adult withficoll solution. CIK cells and DC cells from PBMC were induced with differentcytokines.The proliferation ability of CIK cells was measured by calculation. The phenotypeof CIK and DC cells was analyzed by flow cytometer. We will induce DC with thawedgastric carcinoma antigen. The DC pulsed with gastric carcinoma antigen was coculturedwith the CIK,then Ag-DC-CIK cells was used to kill gastric carcinoma xenografts modifiedby newcastle diease virus.The experiment includes four groups:①Group PBS:PBS+SGC-7901②Group Ag-DC-CIK: Ag-DC-CIK+SGC-7901③Group CIK+NDV:CIK+SGC-7901+NDV.④Group Ag-DC-CIK+NDV: Ag-DC-CIK+ SGC-7901+NDV.The effects of treatment were evaluated by the tumour volumes and necrocytosis area.Results: Soluble antigen can be prepared from gastric cancer cells (SGC-7901) by repetitivefreeze-thaw method, then Ag-DC-CIK cells can be prepared in the presence of the tumorantigen fed DC. The average turnout volumes (cm~3)of NDV+CIK group, Ag-DC-CIK groupand NDV+Ag-DC-CIK group were 1.86±0.27,1.86±0.27 and 0.78±0.19 respectively.There are differents significantly in these groups.(F=48.538, P<0.001) The treatment withNDV+Ag-DC-CIK led a strong suppressive effect on the tumor growth compared withNDV+CIK and Ag-DC-CIK.(P<0.001),but no different between NDV+CIK andAg-DC-CIK.(P=0.862); Turnout inhibition rate of NDV+CIK group, Ag-DC-CIK group andNDV+Ag-DC-CIK group were 29.81%,30.94% and 70.57% respectively.The averagescores of tumour necrocytosis areas in NDV+CIK group, Ag-DC-CIK group andNDV+Ag-DC-CIK group were 1.4,1.6 and 2.4. The differents in these groups aresignificant.(F=11.278, P<0.001) The turnout necrocytosis areas was more than NDV+CIKgroup and Ag-DC-CIK group. (P were 0.011 and 0.035 respectively)There was no differentbetween NDV+CIK and Ag-DC-CIK.(P=0.572)In contrast, treatment withNDV+Ag-DC-CIK led a strong suppressive effect on the tumor growth and more necrocytosis areas.Conclusion: After the xenografts were modified by Newcastle Disease Virus, The Specificcytotoxic Activity of CIK cells induced by dendric cells pulsed with antigen on the gastriccarcinoma Cells was enhanced. The result provides an experimental basis for clinicalapplication as an adoptive immunotherapy.
