| Objective: To investigate the relationship between Endothelia/Endothelia-Areceptor antagonist, nitric oxide/nitric oxide synthase and the blood pressure in renalhypertensive rats, and regulating mechanism of sinitang on the blood pressure.Methods: Total of 24 healthy female Wistar rats were divided randomly into 3groups: renal hypertension group, therapy group and normal control group, with 8 casesin each group. The renal hypertensive models were established by two renal artery clipmethod. After the renal hypertensive models were established, rats in the therapy groupwere treated with sinitang,one time per day for 14 days.Rats in renal hypertension groupwere given saline solution, and ones in the control group were given nothing. The bloodpressure and heart rate were measured with RBF-2 sphygmomanometer. The plasma levelof nitric oxide was determined by nitric reductase colorimetry method, the plasma level ofEndothelia was detected by radio-immunoassay(RIA),and the express of nitric oxidesynthase and endothelia receptor in major target-tissue was detected by immunchistochemical analysis. The statistics analysis was taken with x±SD in quantity data and t testin group compare.Results:①There was no significant difference among the 3 groups before the renalhypertension models were established (P>0.05). The blood pressure was statisticallysignificant different in the renal hypertension group and therapy group before and afterthe establishment of models.②The level of plasma nitric oxide was different in 3 groups,with control group, therapy group and hypertension group from high to low. Endothelia ofthe renal hypertension group decreased, while that of therapy group increased, but with nostatistically significant difference.③The level of ETRA express in target organs of therenal hypertension group was significantly higher than that of the normal control group,while that in renal and heart of the therapy group was significantly lowered than that ofthe renal hypertension group。④The level of endothelial NOS of the renal hypertensiongroup was significantly lowered than that of the normal control group, while that of thetherapy group was significantly higher than that of the renal hypertension groupConclusion: The occurrence of renal hypertension might relate with excess effect ofendothelia and reduce of the level of endothelial NO. Sinitang can effectively relieve the renal hypertension by increasing the expression of endothelial NOS and the level of NOin exterior blood, and decreasing the extra effect of endothelia and the level of ETRA. |
