Objective: To investigate the cytotoxicity of cartap and the the roles of calcium ion and oxidative stress and on its toxic effects in Vero cell line.Methods: 1.Vero cells were incubated in presence of cartap at the concentration of 0.01mmol/L-1mmol/L for 4 h,8 h,12 h,24 h,48h and then the MTT method was used to evaluate the cell viability .At the same time the morphological changes of cultured cells were observed. Vero cells were incubated in presence of cartap at the concentration of 0.01mmol/L-1mmol/L for 8h,12h,24h,48h and then Lactate dehydrogenase (LDH) leakage was measured by colofimetry method to estimate the cell mortality caused by the toxicity of cartap. The fluorescent intensity of ROS was determined by spectrofluorometer after Vero cells treated with cartap at defferent concentrations for 1 h .Vero cells were treated with VitC, verapamil and EGTA at the different concentrition for 30min before incubated in presence of cartap at the different concentration for 24h and then the MTT,LDH was measured.Vero cells were treated with VitC,verapamil and EGTA at the different concentrition for 30min before incubated in presence of cartap at the different concentration for 1 h and then The fluorescence intensity of ROS was measured.Results: Cartap could obviously decrease the viability rate and increase the mortality of Vero cell line in dose-dependent and time-dependent manner. In the meantime ,ROS level in the cell was increased significantly in dose-dependent and manner as well.Conclusions: Cartap has a significant cytotoxicity effect on Vero cell line, and the cytotoxicity mechanism may be that it could induce cellular injury by increasing ROS level and the Calcium has been shown to be an important factor in cell injury resulting.
