At present ,cancer has become a kind of frequently-occurring disease. Being the first or the second reason of death of mankind, it threatens the health of people seriously. In our country, the incidence rate of lung cancer occupies the first place. Because the characteristics of lung cancer are very special, it can not be diagnosed in early-stage easily. Over 75% of the patients were at advanced stage when diagnosed. Therefore many of them were accompanied with Yin-deficiency syndrome, which decreased their immunity, lowered the quality of life of the patients. To effectively solve this problem , we must make sure of the essence of the Yin-deficiency syndrome. Our country has already carried on decedes of years extensive and thorough research in this aspect, but because the essence of Yin-deficiency syndrome was too complicated, up to now we has not a generally acknowledged conclusion yet.Prof. WeiXi Shen, basing on the theories and method of the molecular biology, proposed that the essence of Yin-deficience syndrome can be cytokines, as its pathogensis can be due to the perturbation of cytokine networks characterized by the relation elevation of the gene expression and the biological activity of Interleukin-1(IL-1), Tumour Necrosis Factor(TNF) and so on, which results in the turbulence of cytokine networks.ObjectiveAt present, the theory that the essence of Yin-deficience syndrome is cytokine is only a model of theory. To prove the accuracy of this theory must ask for help of test. In this study, we have used the technology of gene chip and immunohistochemistry to Observe the changing regulations of IL-1α,IL-1β,IL-1R and IL-8, to verify the preliminary conclusion that the pathogenesis of the Yin-deficiency syndrome is due to disequilibrium status of cytokine networks resulted from the relative increase of the bioactivity of the pro-inflammatory cytokine like IL-1, TNF and so on.Methods1. To observe the expression of IL-1a,IL-1β,IL-1R and IL-8 of Yin-deficiency syndrome of lung cancer by immunohistochemistry.We selected 20 cases of lung cancer patients with Yin-deficiency syndrome and that of 20 cases of non-Yin-deficiency syndrome control, They were divided in Yin-deficiency group and non-Yin-deficiency control group. In the Yin-deficiency group, 11 patients are female and 9 patients are male, whose age is from 41 to 70 years old. In the non-Yin-deficiency control group, 10 patients are female and 10 patients are male. They aged from 37 to 70 years old. All the samples were collected from patients who were diagnosed and operated.In this paper, the expression of IL-1a,IL-1β,IL-1R and IL-8 were studied with immunohistochemical method.2. To investigate the gene expression of IL-1a,IL-1β,IL-1R and IL-8 in different cancers with Yin-deficiency syndrome using Affymetrix gene chip.The samples were divided in three groups: 1) lung cancer group including 1 case of lung cancer patient with Yin-deficiency syndrome and 1 case of that without Yin-deficiency syndrome control; 2) gastric cancer group which included 1 case of gastric cancer patient with Yin-deficiency syndrome and 1 case of that without Yin-deficiency syndrome control; 3) colon cancer group including 1 case of colon cancer patient with Yin-deficiency syndrome and 1 case of that without Yin-deficiency a syndrome control. Gene expression profiling was analyzed using AffymetrixResults1. The immunohistochemical results showed: The expression of IL-1a,IL-1βand IL-8 in Yin-deficiency group were Obviously higher than that in non-Yin-deficiency control group(p<0.05). The difference were significant; The difference of expression of IL-1R between Yin-deficiency group and non-Yin-deficiency control group were not significant(p>0.05).2.The Gene Chip results showed: In lung cancer group ,the expression level of IL-1a,IL-1βand IL-8 of Yin-deficiency group is higher than in non-deficiency control group. But the expression level of IL-1R of Yin-deficiency group is lower than in non-Yin-deficiency control group. In colon cancer group,the expression of IL-1a,IL-1β,IL-1R is higher than in non-deficiency control group. But the expression level of IL-8 of Yin-deficiency group is lower than in non-Yin-deficiency control group. In gastric cancer group the expression level of IL-1a,IL-1β,IL-1R and IL-8 of Yin-deficiency group is higher than in non-deficiency control group.Conclusions1. From the result, we found that the IL-1a,IL-1β,IL-1R and IL-8 had some relationship with Yin-deficiency.2. Refering to the result of the Gene Chip ,we propose that the inflammatory cytokines, such as IL-1a,IL-1β,IL-1R and IL-8 maybe the essence and pathogensis of Yin-deficiency syndrome.
