| Heparanase(HPA), an endoglycosidase, releases HS-binding growth factors such as basic fibroblast growth factors (bFGF) and vascularendothelial growth factor (VEGF ), can cleave heparan sulfate (HS) chains in the basement membranes (BM) and extracellular matrices(ECM). It plays an important role in tumor metastasis and become a hot point in tumor diagnosis and therapy research.This article introduced a RNA interference of hepatoma cell which produced a model of heparanase expression inhibition. Flow CytoMeter (FCM), western blot, RT-PCR and immunohistochemistry were used to evaluate this cell model. The result showed that the expression of heparanase in hepatoma cell declined and the hepatoma cell grew slower in nude mice after RNAi.Immunohistochemistry showed positive in the tumor tissue genesis from hepatoma cell and less positive in the tumor tissue genesis from cell after RNAi .This result improved that heparanase could accelerate the growth of tumoer cell and RNAi could inhibit the effection of heparanase. RNAi have a optimistic potential in therapy of tumor.The monoclonal of against the heparanase antibody was produced by hybridoma technique according to the preponderant epitope. The result of western blot showed the monoclonal antibody had a specific affinity for the heparanase and could be used to qualitative analysis the heparanase in the tissues and cells. The monoclonal antibody may be a powerful tool to be used in tumor diagnosis. |
